肾癌中TCF21与KISS-1的表达及调控关系

摘　　要：目的:探讨TCF21与KISS-1在肾癌组织中的表达和相关性以及二者之间的调控关系。方法:实时PCR检测原发未转移肾癌和原发伴转移肾癌组织中TCF21和KISS-1的表达。应用RNA干扰技术沉默Caki-1细胞中TCF21的表达,Western blot检测TCF21和KISS-1蛋白的表达。结果:与原发未转移肾癌相比,原发伴转移肾癌组织中TCF21和KISS-1的表达均显著下调,二者呈显著正相关;转染后TCF21和KISS-1蛋白的表达均显著下调。结论:TCF21和KISS-1与肾癌的侵袭转移相关,TCF21在肾癌细胞中能够正性调节KISS-1的表达。Objective：To research the expression and relationship of TCF21 and KISS-1 in renal cancer,and the regulation relationship between them in renal cancer cells.Methods：Real-time PCR was used to detect TCF21 and KISS-1 mRNA in primary renal cancer without metastasis and primary renal cancer with metastasis.siRNA-TCF21 was transfected to Caki-1 cell,and then the expression of TCF21 and KISS-1 protein was dected by western blot.Results：The expression of TCF21 and KISS-1 mRNA in primary renal cancer with metastasis were much lower than that in primary renal cancer without metastasis,and there was a positive relationship between them.After transfection,the expression of TCF21 and KISS-1 protein was down-regulated significantly.Conclusion：There are relationships between TCF21 and KISS-1 with metastasis of renal cancer,KISS-1 can positively regulate the expression of E-cadherin in renal cancer cells.
