摘  要:目的:探讨我国上海地区汉族人群的对氧磷酶1(PON1)基因T-107C、PON2 Ser311Cys基因多态性与脑梗死发病乃至颈动脉硬化之间的关系。方法:采用聚合酶链反应-限制性片段长度多态性技术,检测134例脑梗死患者和103名健康对照者PON1 T-107C、PON2 Ser311Cys的多态性基因型,并研究其对脑梗死发病的影响。结果:多因素多元逐步Logistic回归分析显示,三酰甘油、总胆固醇是脑梗死的独立危险因素;同时,脑梗死组的PON1 T-107C、PON2 Ser311Cys基因型及等位基因频率分布与健康对照组间差异均有统计学意义(P<0.05),且脑梗死合并颈动脉斑块患者与健康对照组间的PON1 T-107C、PON2 Ser311Cys基因型及等位基因频率分布差异也均有统计学意义(P<0.05)。结论:我国上海地区汉族人群PON1 T-107C、PON2 Ser311Cys这2个位点的基因多态性可能与脑梗死发病乃至颈动脉硬化有关。Objective To investigate the relationship between gene polymorphism of paraoxonase I(PON1) T-107C, PON2 Ser311Cys and cerebral infarction and carotid artery atherosclerosis in Chinese HaM population in Shanghai. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was applied to determine the genotypes of PON1 T-107C, PON2 Ser311Cys in 134 cerebral infarction patients and 103 healthy controls. The impact on the pathogenesis of cerebral infarction was evaluated. Results Multivariate logistic regression analysis showed that triglyceride and total cholesterol were the independent risk factors of cerebral infarction. When compared with normal controls, significant differences were found in distribution of genotypes and allele gene frequencies of PONI T-107C, PON2 Ser311Cys in patients with cerebral infarction (P〈0.05), and a significant difference was also identified in cerebral infarction complicated with carotid atherosclerotic plaque (P〈0.05). Conclusions These findings suggest that PONI T-107C and PON2 Ser31 lCys polymorphism may be associated with the pathogenesis of cerebral infarction, and also with the carotid artery atherosclerosis in Chinese HaM population in Shanghai.
