摘 要:目的 观察CCl4 所致中晚期肝硬化大鼠肠源性内毒素血症的程度及肝组织内毒素的分布。材料与方法 采用鲎三肽基质法测定腹主动脉及门静脉血浆内毒素含量 ;同时 ,采用免疫组化方法对肝硬化组织内毒素的分布进行观察。结果 CCl4 所致肝硬化动物血浆内毒素含量明显高于正常 ,尤以动脉内毒素含量增加显著 ;同时 ,大部分肝脏组织呈内毒素染色阳性 ,内毒素主要分布于变性之肝细胞浆内 ,呈中度阳性染色。结论 CCl4 所致肝硬化伴有明显的肠源性内毒素血症 ,同时内毒素在肝脏内积聚。提示内毒素可直接进入肝细胞内 ,但其进入肝细胞的途径尚需进一步阐明。Objective Gut-originated endotoxemia and distribution of endotoxin in hepatic tissue of cirrhotic rats induced by CCl 4 were investigated. Methods The Distribution of endotoxin in cirrhotic hapatic tissues was studied by the immunohistochemistry,meantime, the severity of gut-originated endotoxemia was evaluated by biochemical methods of limulus amebocyte lysate (LAL). Results There was a moderate positive staining in the cytoplasm of hepatocytes with fatty and bydrolytic degeneration in cirrhotic rats,obviously differing from normal controls. In addition,there was an increased level of endotoxemia in the aorta and portal vein. Conclusion Gut-originated endotoxemia and endotoxin deposition in hepatocytes happen to the cirrhotic rats induced by CCl 4. However, the mechanism of endotoxin into the cytoplasm of hepatocytes in cirrhotic rats is not clarified yet.
