摘  要:目的 探讨硒蛋氨酸对胃癌细胞BGC-803增殖及硒蛋白P表达变化的影响.方法 用不同浓度硒蛋氨酸处理胃癌细胞BGC-803,分别于24、48、72、96小时4个时间点用四甲基偶氮唑盐比色法(MTT法)检测其对细胞增殖的影响,用免疫组织化学SP法观察胃癌细胞BGC-803中硒蛋白P表达的变化.结果 各浓度硒蛋氨酸处理的胃癌细胞BGC-803增殖速度较空白对照组明显减慢(P<0.05),呈浓度、时间依赖性,当硒蛋氨酸浓度为20 mmol/L作用96小时,胃癌细胞BGC-803的生长速度最慢;随着硒蛋氨酸浓度增加,硒蛋白P的阳性表达逐渐增强,且阳性细胞表达率逐渐升高.结论 硒蛋氨酸对胃癌细胞BGC-803增殖具有一定的抑制作用,硒蛋氨酸能增强胃癌细胞BGC-803中硒蛋白P的表达.Objective To investigate the effects of selenomethionine on the proliferation and the expression of selenoproteins P in the gastric cancer cell line BGC-803. Methods Respectively treated' on 4 time points (24,48,72,96 hours) with different concentrations of selenomethionine in gastric cancer cell line BGC-803, the cell proliferation was detected by MT'F assay, the P expression of selenoproteins was detected by immunohistochemistry (SP). Results The MTI" results showed that compared with blank control group, the proliferation of gastric cancer cells BGC-803 treated by various concentrations of selenomethionine were significantly decelerated (P 〈 0.05 ). The deceleration degree depended on the concentration and time, especially after 96 hours in the concentration of 20 mmol/L with Selenomethionine. In ad- dition,with the increase of selenomethionine' s concentration, the positive expression and the positive ex- pression rate of selenoprotein P were gradually increased in gastric cancer ceils BGC-803. Conclusion Selenomethionine can inhibit the proliferation of gastric cancer cells BGC-803 and enhance the expression of selenoprotein P.
