摘 要:为了提高葛根素口服制剂的生物吸收效果,采用研磨法将葛根素与蚕蛹油制成均质混悬剂,采用高效液相色谱法测定小鼠灌胃该混悬剂(葛根素剂量为200 mg/kg)后不同时间点取样血清中葛根素的含量,分析添加蚕蛹油对小鼠口服葛根素生物利用度的影响。试验结果表明,该测定方法的精密度相对标准偏差(RSD)<15%,符合生物样品定量分析方法指导原则的要求;葛根素血清标准品溶液稳定性良好,可用于葛根素生物利用度的分析。建立的药-时曲线数据经DAS软件处理显示:对照组的tmax、Cmax和AUC0~10 h值分别为(35.83±5.48)min、(0.79±0.18)μg/mL、(1.79±0.10)μg.h/mL;试验组的tmax、Cmax和AUC0~10 h值分别为(35.71±4.08)min、(3.72±0.68)μg/mL、(7.00±1.03)μg.h/mL,葛根素的相对生物利用度为391%。根据上述测试与分析结果认为,添加蚕蛹油可以显著提高小鼠对葛根素的口服吸收效果。In order to increase the absorption of puerarin taken orally by organisms,puerarin was ground and mixed with silkworm pupa oil to prepare a homogeneous suspension(equivalent to 200 mg/kg of puerarin) which was used to treat mice intragastrically.High-performance liquid chromatography was employed to determine puerarin content in mouse serum at different time points to evaluate the effect of silkworm pupa oil on bioavailability of orally taken puerarin.The results indicated that the relative standard deviation(RSD) of accuracy was less than 15%,meaning that this method could meet the requirement of guidance for bioanalytical method.The serum standard solution of puerarin was stable and could be used to determine the bioavailability of puerarin.Analysis on parameters of the established pharmacokinetic curves by DAS software displayed that tmax,Cmax and AUC0-10 h values of the control group were(35.83±5.48) min,(0.79±0.18) μg/mL and(1.79±0.10) μg·h/mL,while those of the experimental group were(35.71±4.08) min,(3.72±0.68) μg/mL and(7.00±1.03) μg·h/mL.The relative bioavailability of puerarin was 391% in the experimental group.According to these results,it is concluded that addition of silkworm pupa oil can effectively improve bioavailability of puerarin from oral intake by mouse.
