摘 要:目的探讨ERα/ERβ、TGFβ1蛋白在乳腺癌发展过程中的表达变化。方法收集了2009年1月至2010年12月在山东大学第二医院乳腺外科及临沂市人民医院普外三科入院治疗的258例患者的病理资料,采用免疫组化方法检测正常乳腺组织(n=61)、乳头状瘤(n=65)、乳腺原位癌(n=45)及乳腺浸润性癌(n=87)组织中ERα/β、TGFβ1蛋白的表达情况。所有数据应用方差分析。结果 ERα/ERβ的比值在正常乳腺组织(0.34)、乳头状瘤(0.43)、原位癌(0.68)及浸润性癌细胞(0.71)呈逐步上升的趋势。TGFβ1总体表达情况在乳头状瘤、原位癌及浸润性癌与正常乳腺组织相比差异均有统计学意义(P<0.05),原位癌及浸润性癌强度分数高于乳管内乳头状瘤与正常乳腺组织(P<0.05)。结论在乳腺癌发生发展过程中ERα/ERβ升高,提示ERβ可能是乳腺癌的一个自我保护因素。TGF-β1蛋白表达可能是乳腺癌的早期事件,如能在早期阶段对TGFβ1的表达进行干预,可能对乳腺癌的防治起到重要作用。Objective To detect the expression change of ERα/ERβ and transforming growth factor β1 ( TGFβ1 ) in the development of breast cancer. Methods The pathological data of 258 cases surgically treated in department of breast surgery, the Second Affiliated Hospital of Shandong University and department of general surgery, Linyi People's Hospital from January 2009 to December 2010 were collected. Immunohistochemistl7 were used to detect the expression of ERα/ERβ and TGFβ1 in breast normal tissue (n=61) , papilloma (n= 65) , carcinoma in situ (n =45) and invasive breast cancer ( n = 87) tissues. The analysis of variance was used for data processing. Results ERa/ERβ was 0. 34, 0. 43, 0. 68 and 0. 71 in normal breast tissue, papilloma, carcinoma in situ and invasive breast cancer tissue respectively, indicating the tendency of increasing. The total score of TGFβ1 expression in papilloma, carcinoma in situ and invasive breast cancer were statistically different from that of normal breast tissue (P〈0. 05). The expression intensity score of TGFβ1 in carcinoma in situ and invasive breast cancer were statistically higher than that in papilloma and normal breast cancer tissue ( P〈0. 05 ). Conclusions The ERα/ERβ increased in the development of breast cancer, which hinted that ERβ may play a protective role against carcinogenesis. TGFβ1 protein expression may be an early event in breast cancer, so the intervention on TGFβ1 expression in early stage may have positive effects in breast cancer prevention.
