食管鳞癌患者血清蛋白质谱分析及其与预后的关系

摘　　要：目的用表面增强激光解析离子化飞行时间质谱（SELDI—TQF—MS）仪检测食管鳞癌患者血清蛋白表达水平的差异，并分析其与患者预后的关系。方法用SELDI—TOF—MS仪和弱阳离子纳米磁珠检测144例初诊食管癌患者和50例健康人的血清蛋白，进行比较，并对所有患者进行随访，分析患者血清蛋白的差异与预后的关系。结果144例初诊食管癌患者和50例健康人的血清蛋白比较，在分子量1500～30000范围内，共检测到93个蛋白峰，其中10个差异峰有统计学意义；92例存活期2年以上患者与52例存活期2年以下患者比较，蛋白峰的差异值无统计学意义。Ⅱ期患者中存活期2年以上与存活期2年以下的患者比较，有2个蛋白峰的差异值有统计学意义；Ⅲ期患者中存活期2年以上与存活期2年以下的比较，有1个蛋白峰的差异有统计学意义。结论食管鳞癌患者血清蛋白质谱的差异与患者的预后有一定关系，蛋白指纹图谱检测结果对临床治疗有指导意义。Objective To investigate the correlation of differential expression of serum proteins with progress in patients with esophageal squamous cell carcinoma. Methods The expression of serum proteins was analyzed by surface-enhanced laser desorption/ionization time of flight mass spectrometry （SELDI-TOF-MS） in 144 patients with esophageal squamous cell carcinoma and 50 healthy subjects. The correlation of differential expression of serum proteins and the prognosis of patients were analyzed. Results There were significant difference in expression of 10 serum proteins between cancer patients and healthy subjects. There were no significant difference in protein expression between patients who survived for 〉2y （n=92） and those who survived 〈2y （~=52）. In patients with stage II cancer, the expression of two protein peaks was significant different between pa- tients who survived for 〉2y and patients who survived for 〈2y; while in patients with stage III cancer one protein peak presented significant difference between two groups. Conclusion The differential expression of serum proteins in esophageal squamous cell carcinoma may be correlated with the prognosis of patients.
