摘  要:目的观察旋覆花素对β-淀粉样蛋白25~35片段(β-amyloid protein 25~35,Aβ_(25~35))海马内注射所致阿尔茨海默病(Alzheimers disease,AD)模型大鼠海马Caspase-3表达的影响,探讨其对AD大鼠的治疗作用及机制。方法 30只SD大鼠随机分为对照组、模型组及给药组,Aβ_(25~35)海马内注射制备AD大鼠模型,给药组给予旋覆花素26mg·kg^(-1)·d^(-1)、模型组给予等量溶剂,分2次灌胃,共21d。免疫组织化学及Western blot测定大鼠海马Caspase-3表达。结果模型组海马Caspase-3表达升高,给药组Caspase-3表达下降,与模型组比较差异有统计学意义(P<0.05)。结论旋覆花素主要通过抑制Caspase-3的表达,从而抑制Aβ诱导的AD模型大鼠脑海马的神经细胞凋亡。Objective To explore the molecular mechanism of inulicin in rats with Ahheimers disease (AD). Methods Sprague-Dawley (SD) rats weighing of (364.5 ± 15.0) g were microinjected with β-amyloid protein 25 -35 (Aβ25-35) into the hippocampus to induce AD model rats. In inulicin treatment group, the animals were pretreated with inulicin (contain 1-o-acetylbritannilactone 2.66g/L) dissolved in tween-80 3 days before operation, then inulicin was administered to rats at dose of ABL 26mg · kg-1 . d- 1 by gastric gavage orally for 21 days (n = 10 ). The casepase-3 protein in hippocampus was detected by immunohistochemistry and western blot. Results Casepase-3 expression was more intensive in hippocampus cell of model rats than that of control rats (P 〈 O. 05 ), but there was no signifcant difference between innulicin-treated and control rats (P 〉 0.05). Conclusion Inulicin could inhibit the apoptosis by Aβ injection into hippocampus by down-regulating casepase-3 expression.
