摘 要:目的探讨白细胞介素-24(IL-24)、血管内皮生长因子(VEGF)的表达与肿瘤内微血管密度(IMVD)之间的关系。方法采用免疫组织化学ABC法检测40例脑胶质细胞瘤标本和10例非瘤脑组织中IL-24、VEGF蛋白的表达,以及IMVD、CD34标记肿瘤组织血管内皮细胞。结果高级别胶质瘤的肿瘤微血管密度(MVD)、低级别胶质瘤的MVD、非瘤脑组织的MVD之间比较差异均有统计学意义(P<0.01)。IL-24表达与病理组织学分级呈显著负相关(r=-0.430,P<0.05)。VEGF表达与胶质细胞瘤病理组织学的分级呈显著正相关(r=0.505,P<0.05)。IL-24和VEGF阳性表达与阴性表达的MVD之间比较差异均有统计学意义(P<0.01)。结论 IL-24、VEGF蛋白存在于人脑胶质瘤细胞中,它们与肿瘤血管生成之间具有良好相关性。IL-24、VEGF的表达与IMVD的测定可作为判断胶质细胞瘤恶性程度的重要生物学指标,对判断人脑胶质瘤的临床病理分级具有重要价值。Objective To study the relation between interleukin-24 (IL-24), vascular endothelial growth factor (VEGF) expression and microvessel density(MVD). Methods Expression of IL-24 and VEGF in 40 human glio- ma samples and 10 normal brain tissue was detected by ABC immunohistochemical method. Endothelial cells were marked in neoplasm vessel by CD34 in order to count intratumoral microvessel density (IMVD). Results Signifi- cant difference exists between low and high-grade gliocytoma. It was also noted between the nomal brain tissues and low-grade gliocytoma(P 〈0. 01 ),but no significant difference exists between Ⅰ-grade and Ⅱ-grade or between Ⅲ- grade and Ⅳ-grade gliocytoma. Significant negative correlation ( r = - 0. 430, P 〈 0. 05 ) was found between the IL-24 expression and pathologic histology ratings. Significant positive correlation ( r = 0. 505, P 〈 0. 05 ) was found between the VEGF expression and pathologic histology ratings. Significant negative correlation was found between the IL-24 expressions and angiogenesis in glioma. Significant positive correlation was found between the VEGF ex- pressions and angiogenesis in glioma ( P 〈 0. 01 ). Conclusion IL-24 and VEGF protein exist in cell of human gliocytoma, both of them have close correlation with angiogenesis in tumors. The expressions of IL-24 and VEGF protein, along with IMVD are important biological markers of tumor angiogenesis. United detection of IL-24 and VEGF protein expression could probably play an important role in clinical pathological classification of glioma.
