摘 要:目的研究PI3K/AKT信号通路靶向抑制剂LY294002对食管癌细胞EC109中FR的表达的影响,从而了解食管癌生物学行为与叶酸受体的关系。方法将LY294002作用于食管癌细胞株EC109,四甲基偶氮唑蓝(MTT)法测定细胞增殖抑制率,流式细胞术检测食管癌细胞FR的表达。结果随LY294002浓度的增加及作用时间(12h、24h)的延长,实验组EC109细胞的增殖抑制率逐渐增大,LY294002抑制作用与浓度及作用时间呈正相关。LY294002干预食管癌细胞24小时后,实验组叶酸受体表达的表达较对照组明显降低。结论阻断PI3K/AKT信号通路可以改变食管癌细胞株EC109中FR的表达,并能明显抑制细胞增殖,其机制可能与其抑制AKT磷酸化,从而改变食管癌细胞的生物学行为有关。Objective to study the PI3K/AKT signaling pathway inhibitors targeting LY294002 for EC109 esophageal cancer cells FR expression, the influence of to learn the relationship between esophageal cancer biology behavior and floate receptors. Method LY294002 was used in EC109 esophageal cancer cell lines, tetramethyl azo thiazole blue cell proliferation inhibition rate (determined by MTT method, flow cytometry detection FR expression of esophageal cancer cells. Results With the increase of the LY294002 concentration and action time (12 h, 24 h), the experimental group EC109 cell proliferation inhibition rate increased gradually, LY294002 inhibitory effect was positively correlated with the concentration and action time. LY294002 intervention in esophageal cancer cells after 24 hours, the experimental group expression of folate receptor expression significantly lower than the control group. Conclusion Blocking PI3K/AKT signaling pathway can change FR expression in EC109 esophageal cancer cell lines, and can significantly inhibit cell proliferation, inhibition of AKT phosphorylation, and the mechanism may be to alter the biological behavior of esophageal cancer cells.
