摘 要:目的以缬草药材为研究对象寻找更简单、准确、高效、重现性好、样品用量少的中药材挥发性成分提取与分析联用分析方法。方法采用顶空固相微萃取技术(HS-SPME),分别用100μm PDMS,85μm PA,65μm CW/DVB,65μm PDMS-DVB四种类型萃取纤维头作为考察条件,结合GC对恩施产缬草药材中挥发性成分进行了分析研究;优选缬草药材挥发性成分分析的最佳条件。结果确定最佳萃取分析条件为:缬草药材取样量1 g,在50℃预热30 min,采用85μm PA型Fiber,SPME20 min。结论 SPME分析结果表明,缬草中的主要挥发性成分均能够被有效萃取出来,且色谱图中各色谱峰均具有良好分离度,从而为探索中药缬草的挥发性成分分析提供了一种新方法和新途径。Objective Valeriana officinalis L.is the research object,in order to find an appropriate technique for volatile components extraction and analysis that is easy for application,efficient,accurate and repeatable.Furthermore,it needs very little amount of sample and solvent-free.This will enable us to control the quality of Chinese herbal medicines with reliable and convenient techniques. MethodsHeadspace solid-phase micro-extraction(HS-SPME) combined with GC technology was applied to detect the volatiles of the underground part of the Valeriana officinalis L.plant.A preliminary screening of SPME fibers of various coatings was carried out and the systematically optimized condition for the SPME extraction of the volatiles in Valeriana officinalis L.was obtained. ResultsThe best analysis condition is:85 m PA fiber,1 g plant material,preheating for 30 min and SPME for 20 min after that at 50 ℃. ConclusionExperiment results showed that all the main volatile components can be extracted effectively and all the peaks in chromatogram have shown good resolution,which provided us a novel method for the study of volatile compounds in traditional Chinese medicine Valeriana officinalis L.
