摘 要:目的探讨骨形态发生蛋白-7(BMP-7)对大鼠局灶性脑缺血再灌注损伤后Bcl-xl和半胱氨酸天冬氨酸蛋白酶3(Caspases-3)表达的影响。方法成年健康雄性SD大鼠38只,应用线栓法建立大鼠局灶性脑缺血再灌注损伤模型,随机分为假手术组(A组)、模型组(B组)、BMP-7治疗组(C组),C组于再灌注后立即尾静脉注射BMP-7(0.1mg/kg),A组和B组同步给予等量生理盐水,于再灌注24h后行神经功能缺陷评分,原位末端标记(TUNEL)法检测梗死脑组织细胞凋亡情况,免疫组化方法检测脑组织中Bcl-xl和Caspase-3的表达。结果缺血2h再灌注24h后,与B组相比,C组大鼠神经功能缺陷评分明显降低(t=3.35,P<0.01);TUNEL检测阳性细胞数明显减少(F=409.95,P<0.01);缺血侧脑组织Bcl-xl的表达明显增强(F=229.31,P<0.01),而Caspase-3的表达明显减弱(F=848.38,P<0.01)。结论 BMP-7可能通过增强Bcl-xl的表达和抑制Caspase-3的表达对大鼠局灶性脑缺血再灌注损伤发挥一定的保护作用。Objective To explore the effect of bone morphogenetic protein-? (BMP-7) on the expressions of Bcl-xl and Caspase-3 after focal cerebral ischemia-reperfusion injury in rats. Methods A rat model of focal brain ischemia-reperfusion (I-R) injury was created in 38 adult healthy male SD rats by using suture-occJuded method. The rats were randomized to sham-operation group (group A), model group (group B) and treatment group (group C). The rats in group C were immediately given intravenous BMP-7 (0.1 mg/kg) via caudal vein after I-R injury, and those in groups A and B were given equal volume of normal saline. After reperfusion of 24 hours, the rats' neurological deficit score was recorded, the apoptotic cells were counted by TUNEL-immunoflu- orescence assay, and immunohistochemistry was used to determine the expressions of Bcl-xl and Caspase-3 in brain tissue. Re- sults Compared with group B-after ischemia for 2 hours and I-R 24 hours-the rats' neurological deficit score in group C signifi- cantly declined (t =3.35,P(0.01), the positive cells detected by TUNEL significantly decreased (F=409.95 ,P(0.01), and the expression of Bcl xl in the brain tissue of ischemic side significantly enhanced (F = 229.31, P 〈0.01), but Caspase-3 weakened (F=848.38,P〈0.01). Conclusion BMP-7 exerts its protection against focal cerebral ischemia-reperfusion injury is Iikely via enhancing the expression of Bcl-xl and inhibiting that of Caspase-3.
