摘 要:目的优化制备四逆汤(附子、炙甘草、干姜)的最佳工艺参数。方法以单酯型生物碱、甘草酸及干膏收率为化学指标,以昆明种清洁级小鼠(每组10只)的抗脑缺氧能力(喘气次数和喘息时间)为药效指标,采用L9(34)正交设计试验优选四逆汤的制备工艺;人工神经网络优化模型进一步优化工艺参数。结果四逆汤的最佳制备工艺参数为提取3次,料液比为1∶8,每次提取1.5 h。结论人工神经网络与正交试验联用优选的四逆汤制备工艺参数稳定、可行。AIM To optimize the preparation technology of Sini Decoction(Aconiti lateralis Radix praeparata,Glycyrrhizae Radix et Rhizoma praeparata cum melle,Zingiberis Rhizoma).METHODS The contents of monoester diterpenoid alkaloids,glycyrrhizic acid and yield of dry extract were used as chemical markers.The cerebral anoxia injury,ten mice(clean class) per group,were as the pharmacodynamic indexes.The optimal extraction was selected by orthogonal design L9(34).Then an artificial neural network model was established to obtain the optimum extraction parameters,and proved by experiment.RESULTS The optimal parameters were as follows,3 times,1.5 h every time and the material liquor ratio of 1∶8.CONCLUSION The optimal parameters of Sini Decoction are obtained steadily and efficiently by application of orthogonal design and artificial neural network.
