摘  要:目的用离子色谱法定性分析多花野牡丹精制提取物中亲水性有机酸并定量测定柠檬酸。方法样品稀释液经PT-系列固相萃取柱和0.45μm水膜滤过后,用Ion Pac AS11-HC阴离子分离柱分离,以EG 40淋洗液自动发生器生成的1~40 mm/L KOH溶液梯度洗脱,抑制电导检测器检测。结果除草酸和富马酸未完全分离外,其余组分均可以达到完全分离。柠檬酸质量浓度在5~60μg/mL(r=0.999 9)范围内与峰面积呈良好线性关系,平均加样回收率为100.27%。结论多花野牡丹精制提取物中含有乳酸、乙酸、甲酸、苹果酸、酒石酸、草酸、柠檬酸等亲水性有机酸,其中柠檬酸量高于3%。AIM To qualitatively study organic acids and determine the citric acid content in the refined extract of Melastoma affine by using ion chromatography method.METHODS The diluted sample solution was purified by PT-series solid-phase extraction column and 0.45 μm inorganic membrane,finally separated on an anion column of Ion Pac AS11-HC with the gradient elution of 1-40 mm/L KOH produced by EG 40 eluent automatic generator and determined with a suppressed conductivity detector.RESULTS The remaining organic acids were well separated except oxalic and fumaric acid.Good linear relationship of citric acid existed between the mass concentration and the peak area in the range of 5-60 μg/mL(r=0.999 9).The average recovery was 100.27%.CONCLUTION The refined extract of Melastoma affine contains lactic,acetic,formic,malic,tartaric,oxalic and citric acid.The citric acid content in the sample is over 3%.
