线粒体促凋亡因子Omi/htrA2在结肠癌组织中表达的研究

摘　　要：目的探讨丝氨酸蛋白酶Omi/htrA2在结肠癌组织、癌旁组织、结肠正常黏膜组织中的表达及其临床意义。方法采用免疫组化SP法检测46例结肠癌组织、15例癌旁组织及15例结肠正常黏膜组织中Omi/htrA2的表达,并分析其表达与结肠癌临床病例特征及预后的关系。结果 Omi/htrA2在结肠癌组织中的阳性表达率为69.57%(32/46),高于癌旁组织(13.33%,2/15)和正常结肠黏膜(6.67%,1/15),差异有统计学意义(P<0.01)。结肠癌组织中Omi/htrA2的表达与患者的性别、年龄、及肿瘤大小无关(P>0.05);与肿瘤的分化程度、浸润深度、淋巴结转移和临床分期有关(P<0.05)。结论 Omi/htrA2在结肠癌组织中高表达,其表达与结肠癌分化程度、浸润深度、淋巴结转移及TNM分期有关。结肠癌可能需要Omi/htrA2的表达来促进凋亡,Omi/htrA2的表达对结肠癌的发展有重要作用。Objective To investigate the expression and clinical significance of the protease Omi/htrA2 in colon carcinoma tissue and its association tissues and normal mucous membrace of colon tissues.Methods Omi/htrA2 protein expression levels were detected by immunohistochemistry method in resected colon carcinomas（n=46）,adjacent noncancerous tissues（n=15）,and normal tissues（n=15）,and its association with clinicopathological features and prognosis were analyzed.Results Omi/htrA2 expression was positive in 69.57%（32/46）of colon cancer tissues,which was significantly higher than that in adjacent noncancerous tissues and normal tissues（P0.01）.There were no significant differences in Omi/htrA2 expression with respect to sex,age,and tumor size（all P0.05）.Omi/htrA2 expression level was significantly associated with tumor differentiation,depth of invasion,extent of lymph node metastasis,and tumor stage（all P0.05）.Conclusions Omi/htrA2 expression is more common in colon carcinoma.Omi/htrA2 expression is associated with tumor differentiation,depth of invasion,extent of lymph node metastasis,and tumor stage.Colon carcinoma may need Omi/htrA2 expression for apoptosis,and Omi/htrA2 expression might play an important role in the development of colon carcinoma.
