摘 要:目的探讨肿瘤转移抑制基因kai1在宫颈癌的表达及与人乳头状瘤病毒(HPV)感染的相关性。方法对11例原位癌、46例浸润癌、50例正常宫颈组织石蜡标本分别采取免疫组化法测定kai1蛋白表达,以PCR扩增-琼脂糖凝胶电泳法检测宫颈癌组织中的HPV16E6、E7、HPV18E6/E7情况。结果正常宫颈组织中kai1蛋白阳性表达明显高于原位癌及浸润癌,原位癌亦低于浸润癌,将3组kai1蛋白不同表达强度进行秩和检验,差异有统计学意义(H=8.49,P<0.05),正常宫颈组织明显高于原位癌(H=6.63,P<0.05),原位癌高于浸润癌(H=7.18,P<0.05);HPV16E6、E7、HPV 18E6/E7在原位癌中阳性表达率为63.64%、72.73%、0.91%,在浸润癌中阳性表达率分别为65.22%、54.35%、13.04%,原位癌与浸润癌中阳性表达率比较差异无统计学意义;进行Spearman分析,HPV16E6、E7、HPV 18E6/E7感染率与kai1蛋白阳性表达率之间并无相关性(r=0.13,r=0.11,r=0.15,P>0.05)。结论肿瘤转移抑制基因kai1在宫颈癌的表达呈现下调表达,HPV16E6、E7、HPV18E6/E7的感染不影响kai1蛋白在宫颈癌中的表达。OBJECTIVE To investigate the expression of tumor metastasis suppressor genekail in cervical cancer and analyze its correlation with human papilloma virus (HPV) infections. METHODS Totally 11 eases of carcinoma in situ, 46 cases of infiltrating carcinoma, 50 cases of normal cervical tiesue paraffin specimens were taken respective ly by immunohistoehemieal method for determination of hail protein expression, PCR amplification-agarose gel e lectrophoresis method was employed to detect cervical cancer tissue of the HPV16E6, E7, HPV18 E6/E7 situa- tion. RESULTS The normal cervical tissues kail protein positive expression was obviously higher than that of the carcinoma in situ and infiltrating carcinoma, carcinoma in situ also below the infiltrating carcinoma, the difference in the hail protein expression strength rank and inspection between the three groups was statistically significant (H=8.49, P^0.05), the normal cervical tissue was obviously higher than that of the carcinoma in situ (H=6. 63, P^0.05), carcinoma in situ higher than infiltrating carcinoma (H= 7. 18, P〈0.05); the positive rates of expression of HPV16E6, ET, and HPV 18 E6/E7 in the carcinoma in situ were 63.64%, 72.73%, and 0.91%, respectively, and the positive rates of expression in infiltrating carcinoma were 65.22%, 54.35%, and 13.04%, respectively, the difference in the positive rate of expression between the carcinoma in situ and the infiltrating car- cinoma was not statistically significant. The Spearman analysis indicated that there was no correlation between the infection rate of HPV16E6, ET, or HPV 18 E6/E7 and the positive expression rate of hail (r= 0.13, r=0.11, r =0.15, P〉0.05). CONCLUSION The tumor metastasis suppressor gene hail presents downregulated expres sion, and the HPV16E6, E7, or HPV 18 E6/E7 infection does not influence the expression of hail in cervical cancer.
