荣昌猪C-Fos原癌基因克隆与序列分析

摘  要:为获取荣昌猪原癌基因C-Fos的基因序列及对该基因进行进一步的分析与利用,采集荣昌猪睾丸,提取总RNA,通过反转录-聚合酶链反应(RT-PCR)获得荣昌猪C-Fos基因,TA克隆后测序。结果表明,获得的荣昌猪C-Fos基因全长为1143bp,编码氨基酸381个。将荣昌猪与GenBank中野猪,马,牛,羊,家猫,大熊猫,人,黑猩猩,猕猴,大鼠,小鼠,兔C-Fos基因进行同源性比较,比对结果显示,荣昌猪与野猪的同源性最高,核苷酸序列同源性高达99.48%,氨基酸序列同源性高达99.47%,与其他物种则存在较大种属差异。In order to get C-Fos gene sequence of proto-oncogene in Rongchang pig for analysis and utilization the total RNA was extracted from the testis of Rongchang pigs. We used RT-PCR to get the C-Fos gene and se- quenced it after TA cloning. The length of the obtained C-los gene in Rongchang swine is 1 143 bp, encoding 381 amino acids. Homologous comparisons between C-Fos gene sequences in Sus serofa, Equus caballus , Bos taurus, Ovis aries, Fells catus, Ailuropoda melanoleuca, Homo sapiens, Pan paniscus, Macaca mulatta, Rattus norvegicus, Mus musculus , Oryctolagus of GenBank indicate that the homology is up to 99.47 % between Rongchang swine and wild boar and there are greater species difference between Rongchang pig and other species.
