摘 要:为了摸索出一种简便、快速、无毒,红细胞分散均匀,形态结构清晰,且近乎活体形态的扫描电镜制样方法,分别采用CO2临界点干燥法、自然干燥法、非离心脱水法及设置在无水乙醇里自然沉淀5、15和25 min对正常人红细胞进行处理,最后应用扫描电镜观察红细胞形态.结果显示,CO2临界点干燥法、自然干燥法和非离心脱水法所制备出的红细胞形态基本一致:表面光滑规整,呈双凹圆盘形.在无水乙醇里,自然沉淀5和25 min脱水的红细胞大部分中央凹陷较深,凹陷处表面粗糙,有的红细胞还有开裂现象;而自然沉淀15 min脱水的红细胞不仅分散均匀,而且红细胞中央凹陷较浅,表面较光滑,呈双凹圆盘形.In order to establish a simple, rapid, non-toxic SEM sample preparation method of erythro- cytes with even dispersion, clear morphology and living form, CO2 critical-point drying method, natural drying method, non-centrifugal dehydration method and anhydrous ethanol natural precipitation time for 5, 15 and 25 minutes, were employed to treat blood samples. The results of SEM scanning showed that the morphology of erythrocytes was basically consistent by using the above mentioned methods. The eryth- rocytes were found biconcave with smooth and neat surface. Interestingly, in the anhydrous ethanol, the erythrocytes that were naturally precipitated for 5 and 25 minutes demonstrated deep central depression and rough surface, with cracking phenomenon observed in some of the erythrocytes. However, the eryth- rocytes precipitated for 15 minutes were not only dispersed evenly, but also displayed biconcave shape with smooth surface and shallower central depression.
