摘 要:目的:建立萝卜硫苷(4-甲基亚磺酰丁基硫代葡萄糖苷)的定量测定方法。方法:西兰花籽中的萝卜硫苷经溶剂提取,离子交换柱纯化,采用高效液相色谱法(HPLC)进行测定。色谱柱为Global Chromatography SP-120-5-C18-AP(5μm,4.6mm×200mm),以甲醇:水=1:9(含0.1%甲酸,pH4.0)为流动相,检测波长为229nm,流速为1.0mL/min,柱温为室温。结果:在该色谱条件下萝卜硫苷与其他物质得到了有效的分离,且萝卜硫苷在0.14~2mg/mL范围内与峰面积有良好的线性关系,R=0.9996,检测限为0.08mg/mL,萝卜硫苷的加样回收率为99.45%,RSD为0.77%。结论:建立的方法重复性好、精密度高、回收率较好,可作为西兰花籽提取物中萝卜硫苷的定量测定方法。Objective: To establish the method quantitative determination of gulcoraphanin (4-Methylsufinylbutyl glucosionlate). Methods: The gulcoraphanin was extracted from broccoli seeds by solvent, purified by ion exchange column, then detected by high performance liquid chromatogram (HPLC). The analysis was performed on Global Chromatography SP-120-5-C18-AP column with the methanol- water=l:9 (including 0.1% formic acid, pH4.0) as mobile phase, the detecting wavelength was 229 nm and the flow rate was 1.0 mL/min, the column temperature is room temperature. Results: The results show that the gulcoraphanin with other material got effective separation on this detecting conditions, and the gulcoraphanin had good linear relationship with the range of 0.14-2 mg/mL, R=0.9996, the detection limit was 0.08 mg/mL. The recoveries of gulcoraphanin were 99.45%. Conclusion: The method is good on reproducibility, high in precision, recovery is better, which can be used as the method of quantitative determination of gulcoraphanin in broccoli seeds extracts.
