摘 要:目的探讨整合素连接激酶(integrin-linked kinase,ILK)和垂体肿瘤转化基因(pituitary tumor-transforming gene,PTTG)在卵巢肿瘤组织中的表达和微血管密度(microvessel density,MVD)的关系。方法采用免疫组化EnVision法检测76例恶性卵巢癌、20例交界性卵巢肿瘤、10例良性卵巢肿瘤和10例正常卵巢组织标本中ILK和PTTG的表达,并用CD105标记血管内皮细胞计数MVD。参考Fromowitz方法判定免疫组化染色结果。结果 (1)卵巢癌组织中ILK、PTTG呈高表达,且相关性分析表明ILK与CD105的表达呈正相关(rs=0.321,P<0.05),PTTG与CD105的表达呈正相关(rs=0.65,P<0.05),ILK与PT-TG的表达无相关性(rs=0.141,P>0.05)。(2)ILK、PTTG在卵巢癌组织中的表达与组织学分级、有无腹水、有无远处转移和有无淋巴结转移呈正相关(P<0.05),且这些病理指标在ILK、PTTG均阳性组与任一蛋白表达和无蛋白表达组之间差异有显著性(P<0.05)。(3)ILK、PTTG的表达与卵巢肿瘤组织的MVD呈正相关(P<0.05)。(4)ILK、PTTG高表达是卵巢癌患者生存的影响因素(P<0.05),联合分析表明ILK、PTTG均阳性组的患者生存时间明显低于任一蛋白表达组和无蛋白表达组(P<0.05)。结论 ILK、PTTG在卵巢癌组织中高表达,通过促进微血管形成而在卵巢癌的发生、发展中起重要作用,可作为临床判断卵巢癌生物学行为的分子指标。Purpose To investigate the expression of ILK and FTTG and their relationship with microvessel density (MVD) in ovarian cancers. Methods Immunohistoehemistry was used to detect the expression of ILK and FTTG in tissues of malignant ovarian tumor (76 cases) , borderline ovarian tumor (20 cases), benign ovarian tumor (10 cases) to normal ovarian tissue (10 cases). CD105 was used as a marker of vascular endothelial cell to amount MVD. Fromowitz method was used to determine the results of immunohistochem- istry. Results ILK and PTTG showed high-level expression in ovarian cancer tissue, and the Spearman correlation from rank analysis demonstrated that the ILK expression in ovarian cancer tissue was positively correlated with CD105 expression (rs=0. 321, P 〈 0. 05 ) , PTTG expression was positively correlated with CD105 expression ( r = 0. 65, P 〈 0.05 ) , but there was no correlation between ILK and PTFG expression in ovarian cancer tissue. ILK and PTTG expression in ovarian cancer tissue was positively correlated with histo- logic grade ( Ⅰ -Ⅱ , Ⅲ-Ⅳ ) , ascites, remote metastasis and lymph node metastasis ( P 〈 0. 05 ). When the two proteins were anaIyzed as one group, a significant difference was found between the expression of two and that of only one or none ( P 〈 0.05 ). The expression of ILK and PTTG in ovarian cancer tissue had positive correlation with MVD ( P 〈 0.05 ). Furthermore, the strong expres- sion of ILK and FTTG in ovarian cancer tissues significantly correlated with poor prognosis of ovarian cancer patients ( P 〈 0. 05 ) ; when the two proteins were analyzed at the same time, the survival time in patients with both expression was significantly lower than that with the expression of only one or none. Conclusion The increased expression of ILK and PTTG in ovarian cancer tissue plays a critical role in the occurrence and development of ovarian cancer through promoting the formation of capillaries.
