摘 要:为了解犬瘟热病毒(CDV)在广东地区的分子流行情况及遗传变异规律,本研究从广东省多地采集疑似感染犬瘟热病毒的患病犬的血液、体内组织样品100份,经RT-PCR法检测,共检出90份阳性病料。随机选取10份阳性病料进行CDV H基因扩增,序列分析表明该地区内H基因的同源性为96.6%~99.8%,与参考毒株的H基因同源性为89.9%~97.9%。遗传进化分析发现所检测样品大部分属于亚洲I型毒株,使用软件预测H蛋白的糖基化位点数,发现所有被检测野毒株的糖基化位点数为9个,而疫苗株的为4个或7个。本研究明确广东地区犬瘟热病毒H蛋白基因的变异趋势,为疫苗的选择或开发奠定了基础,为广东地区犬瘟热的防控提供了参考。To investigate the prevalence and the extent genetic diversity of canine distemper virus(CDV) in Guangdong province,a total of 100 blood and tissue samples were collected from many regions in Guangdong province to detect the CDV by RT-PCR method,and 90 samples of them were positive.10 positive samples were randomly selected to amplify CDV H gene.Sequence analysis showed that the nucleotide homology among isolates from Guangdong region achieved 96.6%-99.8% and between the reference strains and isolates was 89.9%-97.9%.Phylogenetic anaysis indicated that all 10 isolates belonged to the Asia-I strain.Using the software to predict the H protein glycosylation site,the glycosylation sites of wild strains were 9,while the number of vaccine strains were 4 or 7.This study proved the variation trend of CDVH gene in Guangdong province which laid the foundation for the selection or development of vaccine,and provided a reference for CDV prevention and treatment in Guangdong province.
