摘  要:通过改变基础发酵培养基中的金属离子和氨基酸种类及浓度,研究其对林肯链霉菌的菌体浓度及发酵液生物效价的影响.单因素实验结果表明,在基础发酵培养基中添加0.02g/L氯化锰、0.02g/L钼酸钠、0.15g/L硫酸镁和0.30g/L硫酸锌,调节初始pH为7.0,按6.7%的接种量接种,于30℃,220rpm的恒温振荡培养箱中培养48h后,再依次添加0.06%谷氨酸、0.10%酪氨酸、0.15%多巴和0.06%甲硫氨酸,再于30℃,220rpm的恒温振荡培养箱中培养5d.菌体浓度可达91%(16 000μg/mL),林肯霉素发酵液生物效价为4 200U/mL.In this work,different types of metal ions and amino acids, as well as their concentrations, were confirmed to have effect on the growth of high-yield strains of Lincoln streptomycete and yield of Lincoln chloramphenicol. The result showed that its most suitable growth condition was: adding 0. 02 g/L MnC12, 0.02 g/L Na2MO4,0.15 g pH was 7.0 and the inoculum was tivating for 48 h, then adding 0. 06 onine in turn,and then at 30 ℃ ,220 the cell concentration could reach 91 4 200 U/mL. MgS04 , 0. 30 7% ,at 30 ℃,220 glutamic acid L ZnSO4 into the basic fermentation medium, the rpm of constant temperature oscillation,after cuid,0. 10% tyrosine,0. 15%dopamine and 0.06%methirpm of constant temperature oscillation.
