摘 要:目的探讨tRNA基因在弱精子症患者中的突变频谱及其与弱精子症之间的相关性。方法根据1999年WHO诊断标准,收集132例弱精子症患者和98例正常人的精液标本。采用聚合酶链反应扩增,序列分析,筛查弱精子症组tRNA^is、tRNA^Ser(AGY)、tRNA^Leu(CUN)基因突变,与正常对照组序列变异比较,分析核苷酸变异位点的进化保守性。结果132例弱精子症患者中19例标本检出9个突变位点,其中tRNA^His。基因突变位点2个,tRNA^Ser(AGY)基因3个,tRNA^Leu(CUN)基因4个。tRNA^Ser(AGY)基因突变在弱精子症组的发生频率(7.58%)高于正常对照组(1.02%),差异有统计学意义(P〈0.05),其中tRNA^Ser(AGY)基因m.12234A〉G突变6例,正常对照组中未检出,两组间差异有统计学意义(P〈0.05)。结论tRNA^Ser(AGY)基因m.12234A〉G突变可能是弱精子症发生的相关因素。Objective The aim of this study is to detect the mutation on mitochondrial tRNA^His, tRNA^Ser(AGY) and tRNA^Leu(CUN) gene in sperms of asthenospermia patients, and investigate tRNA^His, tRNA^Ser(AGY) and tRNA^Leu(CUN) gene mutation frequencies and its correlationship between mutation of the three genes and asthenospermia. Methods 132 asthenospermia cases and 98 control cases were collected under the WHO criterion (1999 Edition). The tRNAHis, tRNA^Ser(AGV) and tRNA^Leu(CON) genes of mtDNA were amplified and sequenced. The sequences were compared with normal sequence to analyze the variants. The nucleotide variation locus evolution conservation was analyzed by bioinformatics tools. Results A to-tal of 9 mutations were found in tRNA^His, tRNA^Ser(AGY) and tRNA^Leu(CUN) genes in 19 patients of 132 and 2 case of 98 control asthenospermia, including 2 mutations in tRNA^His gene, 3 mutations in tRNA^Ser(AGY) gene and 4 mutations in tRNA^Leu(CUN〉 gene. tRNA^Ser(AGY) gene mutation frequencies in asthenospermia (7.58%) was significantly higher than that in controls (1.02%) (P 〈 0.05 ). We found 6 cases with m. 12234A 〉 G mutation in tRNA^Ser(AGY) gene in asthenospermia (6/132), but were not found in controls (0/98), the fre-quencies of m. 12234A 〉 G mutation in asthenospermia was significantly higher than those in controls ( P 〈 0.05). Conclusions In the study, we speculated tRNA^Ser(AGY) gene m. 12234A 〉 G mutation may have some correlation with asthenospermia.
