溶血磷脂酸对宫颈癌细胞形态学影响的体外研究

摘  要:目的观察溶血磷脂酸对宫颈癌细胞株Siha生长及细胞形态的影响。方法①体外培养人宫颈癌细胞株Siha,以终浓度为1μmol/L、5μmol/L、10μmol/L、15μmol/L的溶血磷脂酸和终浓度为5mg/L、10mg/L、20mg/L、40mg/L的多西环素分别干预,并设不加任何药物作为空白对照组。各组均干预72h后进行过碘酸雪夫反应(PAS)染色,用倒置显微镜观察各组宫颈癌细胞的生长及形态变化。②体外培养Siha细胞,用终浓度为10μmol/L的溶血磷脂酸和终浓度为40mg/L的多西环素干预,72h后固定过夜,电镜扫描。结果①溶血磷脂酸组Siha细胞随着溶血磷脂酸浓度升高,癌细胞数量明显增多,细胞异形性及可塑性增加;多西环素处理组随着多西环素浓度增加,细胞数量明显减少,细胞核固缩、碎裂,细胞破坏。②电镜下可见经溶血磷脂酸干预后Siha细胞表面出现大量分泌颗粒,细胞变形伸出细长突起相互连接。多西环素组干预者则细胞数量减少,形态轮廓不清晰,细胞皱缩变形,核碎裂,细胞溶解。结论溶血磷脂酸促进宫颈癌细胞株Siha生长增殖并改变Siha细胞形态,而多西环素则具有相反的抑制作用。[Abstract] Objective To observe the effect of LPA on both the shape and growth of the cervical cancer Siha cells. Methods ②After cervical cancer cells Siha were cultured in vitro,one group of them were intervened with the final concentration of LPA(1 9mol/L,5 μmo|/L, 10 μmol/L and 15 μmol/L) and the other group was intervened with the final concentration of Doxycycline(5 mg/L, 10 mg/L,20 mg/L and 40mg/L). The control group was given no intervention. After 72 hours, all the ceils were stained with PAS. Then the cell shape change and the growth situation were observed under the invert microscope. ②After Siha cells were cultured in vitro, they were intervened with LPA(the final concentration is 10 μmol/L) and Doxycycline (the final concentration is 40 mg/L) ,respectively. After 72 hours,they were fixed overnight,and then scanned with electron microscopy. Results ③Group 1 showed that, with the increase of LPA concentration, the number of the Siha cells increased significantly and the cellular atypia and plasticity also raised. On the contrary, group 2 showed that, with the increasing concentration of Doxycycline, there was a significant reduction in the number of Siha cells. At the same time, the pyknosis and fragmentation of cells nucleus were found and then cells broke into pieces. ④In addition, under the electron microscopy,there were a great quantity of secretory granules appearing on the surface of the cells which were intervened with LPA. Mean-while,the cells processes became slenderer in favour of connecting. Oppositely, the number of the cells which were intervened with Doxycycline reduced massively and the cell outline became unclear. Then the nucleus broke and the cells came to cytolysis after shrinkage and transformation. Conclusion LPA can promote the growth and proliferation of Siha cells with cell morphological changes,while Doxycyline has the inhibitory effect on them.
