摘  要:目的 以锌作为防护因素研究微波对体外培养的猪视网膜神经节细胞的脂质过氧化损伤作用及耐受剂量 .方法 体外培养猪视网膜神经节细胞 ,按微波辐照强度不同分为对照组 ,30 m W· cm- 2 组 ,6 0 m W· cm- 2 组及各辐照剂量加锌组 ,微波理疗机于微波屏蔽室内辐照 1h,分别于辐照后 0和 72 h观察细胞形态变化 ,测定超氧化物歧化酶 (SOD)和丙二醛 (MDA)含量 .结果 微波辐照后 ,细胞有聚集现象 ,部分细胞轴突消失 ,细胞 MDA含量明显增高 ,SO D降低 ,加锌各组光镜下细胞形态变化不明显 ,MDA含量较未加锌组有所恢复 ,SOD活性也增高 ;细胞辐照后继续培养 72 h,30 m W·cm- 2 组 MDA含量降低 ,6 0 m W· cm- 2 组 MDA与辐照组无明显差别 ,加锌后再继续培养 72 h,则两组细胞 MDA均比辐照后降低 ,SOD活性增强 .结论 微波可引起视网膜神经节细胞脂质过氧化损伤 ,锌可提高细胞的抗氧化能力 ,在一定程度上减轻微波对视网膜神经节细胞的过氧化损伤 ,其作用效果与作用时间有关 .AIM To investigate the protective effect of zincon the microwave induced lipid peroxide damage to in vitro cultured pig retinal ganglion cells and the cells' tolerance to microwave. METHODS Pig retinal ganglion cells were cultured in vitro and divided into 3 groups of different radiation intensities (control group, 30 mW·cm -2 group, 60 mW·cm -2 group) and 2 more radiated groups with different dosages of zinc added. The activity of intracellular superoxide dismutase (SOD) and the content of malondia ldehyde (MDA) of each group were assayed after they had been radiated for 1 h in a shielded room by 2450 MHz continuous wave physiotherapy machine. 0 and 72 h after radiation, the morphological variation of cells was observed by optical microscopy and the activity of intracellular SOD and the MDA content were determined.RESULTS After radiation, some cells congregated and their axons disappeared. The content of MDA increased significantly compared with that of the control group and whereas activity of SOD decreased. No obvious change was discovered in the zinc added groups by microscopy. The content of MDA recovered and activity of SOD in the zinc added groups increased compared with that of the radiated groups without zinc added. With cells cultured for 72 h after radiation, the content of MDA of the 30 mW·cm -2 group decreased and there was no significant difference between the MDA content of the 60 mW·cm -2 group and that of the control. MDA decreased whereas SOD increased in zinc added groups after the 72 h culture, compared with that in the radiation groups. CONCLUSION The results of this study indicate that microwave can induce lipid peroxide damage to in vitro cultured retinal ganglion cells and that Zinc can improve antioxidative capacity of cells, and to some extent can remedy microwave induced damage of lipid peroxidation to retinal ganglion cells. Besides, the effect of zinc is related to its action time.
