摘 要:目的研究镁合金AZ31B对兔骨骼肌细胞生物学行为的影响。方法采用浸提液培养兔骨骼肌细胞,倒置显微镜观察细胞生长形态,CCK法研究其1、3、5、7 d的细胞增殖活性,荧光染色研究细胞活力,胞内总蛋白测定研究细胞蛋白表达情况。以钛合金浸提液为对照(对照组),设置镁合金浸提液组(镁合金组)及其pH值调节组。结果 3组骨骼肌细胞贴壁良好,随着培养时间的延长,细胞增殖活性逐渐增加,镁合金组细胞毒性为2级,pH值调节组未见细胞毒性,且可以促进骨骼肌细胞的增殖;荧光染色可见镁合金组存在部分细胞红染,提示镁合金组存在部分细胞毒性;蛋白检测提示镁合金组蛋白含量显著下降,pH值调节组蛋白含量则较正常升高。结论镁合金AZ31B对于兔骨骼肌细胞具有良好的生物相容性,且能促进兔骨骼肌细胞的增殖及蛋白含量的增加,为镁合金AZ31B用于骨科置入材料提供了理论支持。Objective To study the effects of magnesium alloy AZ31B on biological behavior of rabbit skeletal muscle cells in vitro. Methods With titanium as control, the extracts of AZ31B were used to culture the rabbit skeletal muscle cells, and pH value of AZ31B extract were adjusted for the third group to culture ceils. Inverted microscope was used to study the cell growth forms after 1, 3, 5 and 7 days. CCK methods were used to study the proliferative activity, cell staining were used to study cell viability, and BCA analysis kit were used to analyze the total protein of cells. Results Skeletal muscle ceils of three groups had good adhesion, and cell proliferative activity gradually increased with prolonged incubation time. The cell toxicity of magnesium alloy extract group was 2, the pH adjusted group could promote the proliferation of skeletal muscle cells and no cytotoxicity was found. Red dye could be found in the cells of magnesium alloy group through fluorescence, suggesting that there were some cytotoxicity in magnesium alloy group. The protein assay found that protein decreased significantly in magnesium alloy group, but increased in the pH adjusted group. Conclusion Magnesium alloy AZ31B has good biocompatibility for the skeletal muscle ceils, and it can promote skeletal muscle ceils proliferation and increase protein content, which provides a theoretical basis for application of magnesium alloy AZ31B in orthopedic implant materials.
