摘  要:目的 :研究热休克恢复期小鼠肝细胞热休克蛋白 (HSP70 )的表达。方法 :小鼠分别以 4 0、4 2、4 4、4 6℃热休克处理 30 min,恢复 8h;4 6℃热休克处理 30 min,然后分别于热休克后 2~ 72 h取肝 ,以免疫组化方法观察 HSP70的表达 ,并用体视学方法对阳性肝细胞体密度进行定量分析。结果 :4 2、4 4、4 6℃均能诱导肝细胞表达 HSP70 ,以 4 6℃组小鼠阳性肝细胞密度最大 ;4 6℃热休克后 8~ 12 h为 HSP70的合成高峰 (P<0 .0 1) ,72 h后基本降解消失。结论 :HSP70的迅速表达与缓慢降解对维持肝细胞的正常生理功能有重要作用。To study the expression of HSP70 in the mouse hepatocytes during the recovery period of heat shock. Methods: The mice were subjected to heat shock at 40℃, 42℃, 44℃, 46℃ for 30 min respectively, then recovery for 8 h; again subjected to heat shock at 46℃ for 30 min, then the liver tissue were taken at 2~72 h after heat shock. The expression of HSP70 was observed with immunohistochemical methods, and quantitative analysis of the density of the positive hepatocytes was performed by sterological method. Results: Heat shock could induce hepatocytes HSP70 expression at 42℃, 44℃, 46℃ and the maximum density of the positive hepatocytes was at 46℃(P<0.01); the synthesis of HSP70 reached the maximum value 8~12 h after the heat shock at 46℃(P<0.01) and its degradation almost disappeared after 72 h. Conclusion: The quick expression and the slow degradation of HSP70 may play an important role in maintaining the normal physiological function of the hepatocytes.
