摘 要:目的 :为高强度超声生物学焦域大体观测提供一种准确可靠、直观、快捷的方法。方法 :采用高强度超声样机对离体子宫和在体移植性兔宫体 VX2 肿瘤进行辐照 ,辐照后即刻肉眼直接观测焦域形态、位置及大小并与氯化三苯基四氮唑 (简称 TTC)染色后未着色区进行比较 ,同时对 TTC染色后的着色区及未着色区组织进行光电镜检查。结果 :(1 )肉眼直接观测到焦域的标本在 TTC染色后均出现了明确的未着色区 ,焦域形态、位置、大小与 TTC染色后未着色区一致 ;(2 )组织学检查发现 TTC染色后未着色区细胞呈凝固性坏死 ,而周围着色区细胞形态正常 ,二者有明确的分界 ;(3) TTC染色未着色区出现率在离体子宫为 1 81 / 1 96 (92 .3% ) ,在体兔宫体肿瘤为 1 0 0 % (40 /40 ) ,明显较肉眼直接观测焦域的检出率 6 6 .8% (1 31 / 1 96 ) ,37.5 % (1 5 / 40 )为高 (P<0 .0 0 5 )。结论 :TTC染色方法对高强度超声生物学焦域凝固性坏死的观测准确可靠、直观快捷 。Objective:To establish an accurate and simple method for the observation of the bilogical focal field of high intensity ultrasound(HIU) Methods:In vitro human uterus and in vivo rabbit VX 2 neoplasm were irradiated with HIU The biological focal field were measured before and after Triphenyl tetrazolium chloride(TTC)staining Then the uterine specimens were studied by scanning electron microscopy or HE staining to evaluate the tissue damage Results:(1)all specimens with macroscopically visible focal field revealed unstained areas after TTC staining The shape,position and size of the focal field correlated well with the TTC unstained araes;(2)histological examnition showed coagulated necrosis in the TTC unstained field with precisely defined margin between the necrotic and TTC stained normal tissue;(3)For in vitro uterus,the positive rate with TTC staining was significantly higher than that observed without TTC staining(92 3% vs 66 8%, P <0 005) For in vivo rabbit uterine neoplasm,the positive rate with TTC staining was 100%,while that without TTC staining was only 37 5%, P <0 005 Conclusion:TTC stain was simple and accurate in the observation of bilogical focal field of HIU,it yielded higher positive rate than that observed macroscopically
