摘  要:目的定量分析甲状腺激素缺乏对围生期大鼠脑GoαmRNA的影响。 方法以PTU溶液灌胃造成大鼠围生期甲减模型 ,部分甲减仔鼠每天腹腔注射T4 2 μg/ 1 0 0 g体重。收集 5、1 5、2 5d的正常、甲减及T4 -替代仔鼠脑组织 ,用竞争性RT -PCR法测定脑GoαmRNA。 结果生后 5、1 5、2 5d的甲减仔鼠脑GoαmRNA水平均较同日龄正常仔鼠明显升高 ,尤以 1 5d时最为显著。T4 -替代可明显减轻这种变化。 结论甲状腺激素对发育期鼠脑Goα基因的表达有下调作用 。Objective To study the effects of thyroid hormone deficiency on the expression of α-subunit of Go in the developing rat brain. Methods Perinatal hypothyroidism was induced by the administeration of propylthiouracil (PTU) solution to the dams by gavage. Some pups were injected intraperitoneally with 2μg T 4/100g BW daily from the day of birth. Brains were collected from the control and the hypo-and T 4-injected hypothyroid rats on postnatal day 5, 15 and 25. Quantification of Goα mRNA in the developing rat brain was performed by competitive RT-PCR using internal and external standardisation. Results Brain Goα mRNA levels in the hypothyroid pups were higher than the agematched controls, and the changes were most marked at 15 days post-partum. The mRNA levels in the T 4-injected pups were significantly lower in relation to the age-matched hypothyroid pups. Conclusion Thyroid hormones may down-regulate Goα expression in the developing rat brain. This may contribute to the actions of thyroid hormones on brain development.
