摘  要:目的:建立一种利用高效液相色谱法同时测定可食药用葱属植物中18种游离氨基酸含量的方法。方法:以异硫氰酸苯酯为柱前衍生化试剂,采用梯度洗脱,于254 nm波长处检测18种氨基酸。结果:在一定的浓度范围内,各种氨基酸峰面积和浓度呈良好线性关系,r值均大于0.998;加样回收率为91.5%~110.5%(n=9)。结论:应用该法对大蒜、洋葱、韭菜等几种常见可食药用葱属植物中18种游离氨基酸的含量进行了测定,准确、方便、快速,可用于不同葱属植物的质量控制研究。Objective:To establish an HPLC method for simultaneous determination of 18 kinds of free amino acids in edible and medicinal allium plants. Methods: The phenyl isothiocyanate was used as pre - column derivatization reagent. A total of 18 kinds of amino acids were detected using gradient elution at 254 nm wavelength. Results:In a certain concentration range, the peak area and concentration of each amino acid showed a good linear relationship. Correlation coefficient r values were more than 0. 998. The recoveries ranged in 91.5% -110. 5% (n = 9). Conclusion:Totally 18 kinds of free amino acids were determined in garlic, onions, leeks and other several common edible and medicinal Allium plants. The method established is accurate, simple and rapid, which is suitable for the quality control of different Allium plants.
