摘 要:雷公藤甲素(Triptolide,简称甲素)是一种二萜内酯类化合物,具有多种生物学活性。准确定位甲素各种药理作用相应的靶分子,不仅有利于揭示其药理作用机制,也可以为其分子结构改进相关研究提供理论基础,最终达到减毒和增效的目的。因而甲素药理作用靶分子的筛选和鉴定成为研究的重点,并在近年来不断取得突破。本文就近年来对甲素药理作用靶分子相关实验研究作一综述,为相关研究人员和临床工作者提供参考。Thunder God Vine, also known as Tripterygium wilfordii Hook F, is a traditional Chinese medicinal plant used for treating a wide variety of diseases from rheumatoid arthritis to chronic nephritis for centuries. Triptolide is a structurally unique diterpene triepoxide isolated from Thunder God Vine. It also possesses a unique profile of biological activities, including anti-inflammatory, anti-rheumatic, immunosuppressive, and antitumor activities. However, owing to its severe hepatotoxicity and renal toxicity, triptolide cannot be used systemically in the clinic. Screening and indentifying the molecular targets of triptolide are of great importance, since these works can lay the base for improved therapeutic potential and lowered toxicity through structural modifications. This paper reviews advances in experimental studies on identification of the molecular targets of triptolide in recent years, which may yield important insights for its clinical application.
