携sialyl lewis^x配基的靶向脂质微泡的构建及体外寻靶的试验研究

摘　　要：目的构建携sialyl lewis^x配基的靶向脂质超声微泡,体外评价其靶向黏附性能。方法采用"生物素-亲和素"桥连法制备携sialyl lewis^x配基的靶向超声微泡(MBs),检测该微泡的一般特性,观察配基与微泡结合情况。检测刺激后血管内皮细胞p选择素的表达量。普通光镜下观察体外靶向结果。结果 sialyl lewis^x配基能与脂质微泡高效连接,结合率为93.93%,微泡大小较均一,平均粒径为1.34μm;激光共聚焦荧光显微镜见细胞胞浆及胞膜大量表达p选择素,且与靶向造影剂高效、特异性结合。结论成功构建出携sialyl lewis^x配基的靶向脂质超声微泡,此靶向声学造影剂在体外具有较强的高效、特异性靶向黏附能力。Objective To prepare a kind of targeted lipid ultrasound mierobubbles binding with sialyl lewisx genin and investigate its affinity in vitro. Methods The lipid ultrasound mierobubbles carried with sialyi lewisx were pre- pared using ＂biotin avidin＂ bridging method. The characteristics of targeted mierobubbles and the combination were valued. The stimulated endothelial cell of p selectin expression and the targeted effectiveness were observed by fluores cenee microscopy. Results Ligands with microbubbles had an efficient connection and the rate was 93.93%. The mi- crobubbles size was uniform, average particle size of 1.34 /~m. Mass of p-selectin expressed on the surface of cell and in the cytoplasm. Combination was efficiency and specific. Conclusions Targeted mierobubbles were prepared suc- cessfully and can bind to cell effectively in vitro.
