摘  要:目的研究肿瘤坏死因子受体相关因子6(TRAF6)在肿瘤恶病质骨骼肌炎症反应中的作用。方法小鼠结肠腺癌细胞接种BALB/c小鼠诱导肿瘤恶病质模型,每天监测各组小鼠体质量,于第7、13和19天检测左侧腓肠肌组织的质量、纤维横切面积,PCR和Western blotting检测TNF-α、IL-6、TRAF6 mRNA和蛋白表达水平。结果与对照组比较,随着接种细胞时间的延长,荷瘤小鼠体质量和去瘤体质量减轻(P<0.05);腓肠肌组织的质量减轻(P<0.05),纤维横切面积减小(P<0.05),TNF-α、IL-6和TRAF6的mRNA和蛋白水平均显著升高(P<0.05),且TRAF6的水平与腓肠肌的消耗呈正相关(P<0.05),与TNF-α和IL-6的水平正相关(P<0.05)。结论 TRAF6可能在肿瘤恶病质骨骼肌组织的炎症反应过程中起到重要的作用。[Objective] To study the role of TRAF6 in cancer cachexia skeletal muscle inflammation. [Methods] Murine colon 26 adenocarcinoma cells were inoculated in male BALB/c mice as models of cancer cachexia. Body weight was detected daily. The left gastrocnemius muscle weight, fiber crosscut area, levels of TNF-α and IL-6, mRNA and protein levels of TRAF6 were detected on day 7, 13 and 19. [Results] When compared with control group, body weight, non-tumor body weight, fiber crosscut area and weight of gastroc- nemius muscle were obviously decreased (P 〈0.05), whereas levels of TNF-αand IL-6, mRNA and protein lev- els of TRAF6 were obviously increased after tumor inoculation (P〈0.05). The level of TRAF6 and the con- sumption of gastrocnemius muscle was positively related (P 〈0.05), whereas the level of TRAF6, TNF-α and IL-6 were positively related (P〈0.05). [Conclusion] TRAF6 may play an important role in cancer cachexia skeletal muscle inflammation.
