摘 要:目的探讨细胞周期蛋白CyclinD1和S期激酶相关蛋白2(Skp-2)在鼻咽癌及鼻咽癌前病变组织中的表达活性及其病理意义。方法鼻咽分化型癌20例、鼻咽未分化型癌20例,鼻咽部癌前病变20例,应用免疫组化方法检测CyclinD1和Skp-2蛋白在标本中的表达活性,比较组间差异。结果鼻咽分化型癌、鼻咽未分化型癌和鼻咽癌前病变组织中,CyclinD1的平均阳性表达率分别为75%、73%和47%,平均阳性表达值分别为2.11±0.43、1.81±0.68、1.15±0.56,分化型鼻咽癌与癌前病变、未分化型鼻咽癌与癌前病变相比较,差异有统计学(P<0.05);Skp-2的平均阳性表达率分别为32%、41%、33%,平均阳性表达值分别为0.53±0.49、0.82±0.54、0.72±0.43。在分化型鼻咽癌,CyclinD1与Skp-2阳性表达值具有显著相关性。结论 CyclinD1全程均参与了鼻咽癌发生、发展、演进过程;Skp-2参与的细胞周期调节可效应能是鼻咽癌发生的早期事件。ObjectiveTo investigate the expressive activities of CyclinD1 and Skp-2 in tissue samples of nasopharyngeal carcinoma (NPC) and precancerous lesion in the nasopharynx and to explore their implications in nasopharyngeai carcinogenesis. Methods Immunohistochemical methods were taken to detect the expressive activities of CyclinD1 and Skp-2 in NPC tissue samples taken from 20 cases of differentiated NPC, 20 cases of undifferentiated NPC, and 20 cases of nasopharyngeal precancerous lesion. Then, a comparative analysis was made among them to see their differences in expressive activities and their possible pathological association with nasopharyngeal carcinogenesis. Results The positive expressive rates of CyclinD1 were 75%, 73% and 47% in tissue samples of differentiated NPC, undifferentiated NPC and nasopharyngeal precancerous lesion, and their averaged expressive values were 2.11+0.43, 1.81+0.68 and 1.15+0.56 respectively, with statistical significance in their differences when compared between differentiated NPC with precancerous lesion and undifferentiated NPC with precancerous lesion (P〈0.05). The positive expressive rates of Skp-2 were 32%, 41%, 33% in tissue samples of differentiated NPC, undifferentiated NPC and precancerous lesion, and their averaged expressive values were 0.53+0.49, 0.82+0.54, 0.72+0.43 respectively, with no statistical significance among them. However, there was statistical significance present in the correlation of the expressive activity of CyclinD1 with that of Skp-2 in differentiated NPC. Conclusion CyclinD1 may be involved in the whole process of nasopharyngeal carcinogenesis begun from the initiation, development until progressing of human nasopharyngeal carcinoma, and Skp-2 may hold such an effect on the regulation of cellular cycle as a earlier event associated with the initiation of NPC.
