摘  要:目的探讨在海拔4300 m高原条件下西地那非对大鼠心肌的影响。方法 Wistar雄性大鼠30只,分为A组(空白对照组)、B组(高原对照组)及C组(高原西地那非组)。在各处标准饲养3 d后处死取心脏,用比色法测定丙二醛(MDA)、超氧化物歧化酶(SOD),取血浆测定肌酸激酶(CK)。HE染色法观察左心室病理结构的改变并评价心肌损伤程度。结果 MDA、CK含量B组较A组明显增加(P<0.05),C组较B组明显减少(P<0.05),SOD含量B组较A组明显减少(P<0.05),C组较B组明显增加(P<0.05)。心肌损伤评分B、C组高于A组(P<0.05),C组与B组相比,损伤明显减轻(P<0.05)。结论在海拔4300 m缺氧条件下,大鼠心肌确实形成了氧化损伤,给予西地那非后能显著改善损伤,对心肌有保护作用。Objective To investigate influence of sildenafil on cardiomyocyte damage at 4300 m altitude in rats. Methods A total of 30 Wistar male rats were randomly divided into group A ( control group) , group B ( high altitude group) and group C (Sildenafi group). All the rats were killed after standard feed for three days, the concentration of Malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in cardiac muscle, and the concentration of creatine kinase (CK) in plasma were measured by colorimetric method. Pathologic and structural changes of left ventricle were observed by HE staining method, and myocardial damage degrees were also evaluated. Results Contents of MDA and CK in group B were significantly increased than those in group A (P 〈 0. 05 ) , while the contents in group C were significantly decreased than those in group B (P 〈 0.05). Content of SOD in group B was significantly decreased than that in group A (P 〈 0. 05 ) , and the content in group C was significantly increased than that in group B ( P 〈 0. 05 ). Scores of myocardial damage in groups B and C were significantly higher than that in group A ( P 〈 0.05 ) , and the score in group C was significantly lower than that in group B ( P 〈 0. 05 ). Conclusion Oxidative damage can be found in car- diomyocyte damage at 4300 m altitude in rats. Sildenafil may improve the injuries and protect cardiac muscle.
