摘 要:优化大叶南五味子干燥果实粗多糖(KPS)的纯化工艺。用三氯醋酸(TCA)法、Sevage法脱蛋白,单因素和正交试验法优化活性炭脱色工艺参数,比色法测定蛋白含量和脱色率。TCA法脱蛋白率达到72.6%,极显著高于Sevage法(P<0.01);TCA法多糖损失率显著低于Sevage法(P<0.05);活性炭脱色的最佳工艺参数:脱色温度60℃,脱色时间80min,溶液pH1,活性炭用量1.5%,脱色率达到86.83%±3.93%(n=6),多糖含量达到19.39%±7.21%(n=6)。纯化KPS粗品最优工艺是用TCA法除蛋白质,1.5%的活性炭脱色。To optimize purification technology of crude kadsura polysaccharides (KPS) which were extracted from dried Kadsura marmorata fruits. The crude KPS were removed protein by trichloroacetic acid (TCA) method and Sevage method, respectively. Based on the single factor tests, the process parameters (decoloring temperature, decoloring time, solvent pH value and active carbon concentration) of active carbon decoloring method were optimized by the orthogonal test. The contents of protein and decoloring ratio were detected by eolorimetric method. The results showed that the ratio of removing protein reached to 72.6% by TCA method and was significantly higher than Sevage method (P〈0.01). And polysaccharide loss rate of TCA method was significantly lower than that of Sevage method (P〈0.05). The optimal technological parameters of active carbon decoloring method was 60℃ of decoloring temperature, 80 rain of decoloring time, pH 1.0 and 1.5% of active carbon concentration. Under this condition, decoloring ratio reached to 86.83% ±3.93%, the content of polysaccharide reached to 9.39%±7.21%. The best purification technology of crude kadsura polysaccharides (KPS) was TCA method removed protein and 1.5% active carbon decoloring.
