摘 要:目的研究Caspase-3蛋白在肌萎缩脊髓侧索硬化症(ALS)转基因模型鼠脊髓不同发病时期的表达变化,探讨ALS发病过程中细胞凋亡情况。方法利用免疫组织化学(SP二步法)方法检测Caspase-3在ALS转基因模型鼠脊髓的发病前期(95d)、发病中期(108d)、发病后期(122d)表达变化,并与同窝野生型鼠(WT)比较。结果 ALS转基因模型鼠脊髓中Caspase-3阳性细胞在发病前期(95d)变化不明显,与同窝野生型鼠比较,差异无显著性;发病中期(108d)Caspase-3阳性细胞明显增多,与同窝野生型鼠比较,差异具有统计学意义;发病后期(122d)Caspase-3阳性细胞显著增多,但较发病前期有所下降,与同窝野生型鼠比较,差异具有统计学意义。结论 Caspase-3阳性细胞在ALS发病不同时期发生变化,表明细胞凋亡在ALS的发生发展中发挥着重要作用。Objective To study the change of expression of Caspase-3 in the spinal cord of ALS transgenic mice at different stage and to diseuss the role of cell apnptosis in the process of ALS disease. Methods The change of expression of Caspase-3 in the spinal cord of ALS transgenie mice was detected by immunohistochemistry ( SP two-step) at the pre-symptomatic(95d) and the symptomatic phase(108d) and the end-stage(122d) ,compared with wild-type mice. Results In the spinal cord of ALS transgenie mice, the change of expression of Caspase-3 at pre-symptomatic (95d} was not obviously and had no significant difference compared with wild-type mice. The Caspase-3 positive cells were increased significantly at the symptomatic phase(108d) and the difference was statistically significant compared with wiht-type mice. The expression was still higher at the end-stage( 122d} and the difference was statistically signifi- cant but lower than the symptomatic phase(108d) compared with wild-type mice. Conclusion The change of Caspase- 3 positive cells at different stage shows that cell apoptosis plays an important role in the development of ALS.
