摘  要:目的:探讨模拟失重后高+Gx暴露对猴牙龈组织诱导型一氧化氮合酶(iNOS)、环氧合酶-2(COX-2)表达的影响。方法:20只雄性猕猴随机分为4组,即正常对照组(A)、30 d模拟失重组(B),+13Gx/230 s组(C)、30 d模拟失重后超重组(D),其中D组又分为+11Gx/270 s(D1)、+13Gx/230 s(D2)和+15Gx/200 s(D3)。相应模拟实验后,放血处死各组动物并取牙龈组织,用免疫组化SABC法分别观察各组猴牙龈组织中iNOS、COX-2的表达。结果:各实验组iNOS、COX-2表达水平均高于对照组(P<0.05)。各组中iNOS的表达水平依次为C组>B组>D1组>D2组>D3组>A组,其中B、C、D1、D2各组间两两相比均无统计学差异(P>0.05),而分别与D3组相比,均P<0.05;COX-2的表达水平依次为D3组>D1组>D2组>C组>B组>A组,其中B、C、D1、D2各组分别与D3组相比P<0.05,B组与C组、D1组与D2组相比均P>0.05,而D1和D2分别与B组、C组相比,均P<0.05。结论:30 d模拟失重和+13Gx/230 s均可引起猴牙龈组织iNOS、COX-2的表达;30 d模拟失重后再高+Gx可使COX-2的表达上调,iNOS的表达下调。AIM: To study possible influence of high + Gx exposure after 30 - day weightlessness simula- tion on iNOS and COX-2 expression in gingiva of rhesus macaque. METHODS:20 male rhesus macaques were ran- domly divided into 4 groups: control group (A), 30 d weightlessness simulation group (B), + 13 Gx group (C) and high + Gx exposure after 30 d weightlessness simulation group (D). The animals in group D were divided into 3 sub- groups and were exposed to + 11Gx/270s ( D1 ), + 13Gx/230s (D2) and + 15Gx/200s (D3) respectively. The gin- giva tissues were obtained and examined histologicaly. The expression of iNOS and COX-2 was detected by immunohis- tochemieal method. RESULTS:The expression of iNOS and COX -2 in gingival tissues in experimental groups was higher than in the control(P〈0.05) and in the order of C 〉B〉D1 〉D2 〉D3 〉A. And there was no significant difference among the group B, C, D I and D2 (P 〉 0.05). The expression of COX-2 was in the order of D3 〉 D1 〉 D2〉 C 〉 B 〉 A. There was a significant difference between group group D3 and B, C, D1 or D2 (P 〈 0.05). CONCLU- SION :30 day weightlessness simulation and high + Gx may induce an increase of iNOS and COX-2 expression in the gingiva. COX-2 reaction may be aggravated after + Gx exposure following 30-day weightlessness simulation, but iNOS -2 expression may be reduced.
