摘 要:【目的】研究陇东地方牛和西门塔尔杂交肉牛2个基因的多态性及与肉质性状的相关性。【方法】采用PCR-RFLP方法对陇东地方牛和西门塔尔杂交肉牛的M-calpain基因和H-FABP基因进行多态性分析,并利用最小二乘法拟合线性模型分析了基因型与肉质性状之间的相关性。【结果】发现M-calpain基因突变位点C1795T和H-FABP基因序列中的突变位点C1006G,分别检测到3种基因型AA/AB/BB和GG/GC/CC。相关分析表明M-cal-pain基因中AB基因型在剪切力上差异显著高于AA基因型(P?0.05);BB基因型的滴水损失显著高于AB基因型(P?0.05);H-FABP基因中GC和CC基因型对牛肉滴水损失有显著的影响(P?0.05)。【结论】推测M-cal-pain基因C1795T和H-FABP基因C1006G的突变位点可以作为陇东地方牛和西门塔尔杂交肉牛牛肉嫩度和大理石花纹的遗传标记。[Objective] Analysis on polymorphism of M-calpain and H-FABP genes in Longdong local cattle and Simmental crossbred beef and its relationship with meat quality trait. [Method] M-call)ain and H-FABP genes were identified with PCR-RFLP analysis in local cattle. The association between genotype markers and meat traits were analyzed by least squares linear model at the level of DNA. [Results] The site C1795T of M-calpain gene and C1006G of H-FABP gene were detected. Three genotypes, including AA/AB/BB and GG/GC/CC, of C1795T and C1006G were found, respectively. The AB genotype of M-calpain gene showed a highly significant effect on shear force than that of AA (P〈0.05). The BB genotype of M-calpain gene had a highly significant effect on retort cooking loss percentage than that of AB (P〈0.05) ; In addition, GG and CC genotype of H-FABP gene had a highly significant effect on retort cooking loss percentage (P 〈0.05). [Conclusion] The site C1795T of M-calpain gene and C1006G of H-FABP gene could be used as genetic markers for beef tenderness and marbling score in Longdong local cattle and Simmental crossbred beef .
