人源抗肿瘤坏死因子-α噬菌体抗体的基因结构分析

摘　　要：从混合的噬菌体抗体库中筛选到了抗人TNFα的人源单克隆抗体 ,并对筛选到的抗体基因进行了序列测定和分析。结果表明 ,筛选到的 4个特异性抗TNFα噬菌体抗体克隆的重链基因序列相同 ,该重链基因长 6 81bp ,编码 2 2 7个氨基酸残基 ,属于人免疫球蛋白第Ⅲ家族 ,其中 1～ 119位氨基酸残基为重链可变区 (VH) ,12 0～ 2 2 7位为重链恒定区 1(CH1)。 4个噬菌体抗体克隆的轻链均缺失 ,因此实际上筛选到的是单重链抗体。Human anti?TNFα monoclonal antibodies were selected from pooled human antibody combinatorial libraries using phage display technology, and their genes were sequenced and analyzed. The results showed that the genes encoding heavy chains of the four anti?TNFα antibodies were the same, which comprised 681 nucleotides and encoded a protein of 217 amino acids, belonging to human IgGⅢ gene family, wherein 1?119 amino acids constituted the V region of the heavy chain (VH) and 120?227 amino acids constituted the C1 region of the heavy chain (CH1). All of the four antibodies lacked the light chain, showing that they were all the same mono heavy chain antibody. The characteristics of heavy chain protein encoded by the heavy chain gene were also analyzed.
