蛋白激酶CK2α在食管癌中的表达及意义

摘　　要：目的研究蛋白激酶CK2α与食管癌发生发展及临床病理因素的相关性。方法采用免疫组织化学方法检测组织芯片中56对食管癌及其癌旁正常黏膜中CK2α蛋白的表达。分别提取9对手术切除的食管癌及其癌旁组织标本的RNA与蛋白,通过qRT-PCR和Western blot的方法分别在基因与蛋白水平检测CK2α的表达水平。结果在56对食管癌及癌旁组织中,CK2α在癌组织中的表达明显强于癌旁正常黏膜组织,在食管癌组织中弱阳性表达率为8.9%,中阳性表达率为35.7%,强阳性表达率为55.4%,而癌旁正常黏膜组织分别为96.4%、3.6%和0.0%,两者差异有统计学意义(P<0.001)。CK2α蛋白在食管癌中的表达水平与其TNM分期有关(P<0.001)。qRT-PCR及Western blot证实无论是在基因水平还是蛋白水平,蛋白激酶CK2α在食管癌组织中均呈高表达。结论蛋白激酶CK2α可以作为食管癌发生发展及判断其进展程度的分子靶标。Objective To investigate the correlation between protein kinase CK2α and the clinicopathological parameters of esophageal carcinoma.Methods Tissue microarrays for 56 esophageal carcinomas and the surrounding normal tissue were used to assess the expression of CK2α by immunohistochemistry.RNA and protein extracted from 9 surgically resected esophageal carcinomas and adjacent tissue specimens were used to assess the CK2α expression via qRT-PCR and Western blot.Results The expression of CK2α was up-regulated in esophageal carcinomas（weak positive 8.9%,medium positive 35.7%,and strong positive 55.4%） compared with that in normal tissues（weak positive 96.4%,medium positive 3.6%,and strong positive 0.0%）.A significant correlation was found between the CK2α expression and the TNM stage of esophageal carcinoma（P0.00）.The RNA and protein expression of CK2α were also higher in esophageal carcinoma than that in normal tissue.Conclusion CK2α may serve as a marker predicting the development and progression of esophageal carcinoma.
