摘  要:目的观察内毒素诱导大鼠心肌损伤时EPO及EPO受体表达。方法 20只雄性SD大鼠随机分为LPS组和对照组,LPS组腹腔注射生理盐水,连续处理3 d后腹腔注射LPS 10 mg/kg,建立内毒素心肌损伤模型。对照组与LPS组腹腔注射等量生理盐水。腹腔注射6 h后于鼠尾部放血测定心肌损伤标志物,断颈活杀取出心脏组织测定EPO及EPO受体浓度。结果 LPS组血清心肌损伤标志物(cTnⅠ、CK、CK-MB)水平均显著高于对照组(t=6.751、5.346、3.432,P<0.05)。LPS组心脏组织EPO及EPO受体水平均显著高于对照组(t=2.587、4.306,P<0.05)。结论内毒素诱导大鼠心肌损伤时存在EPO及EPO受体高表达,为外原性EPO在脓毒症的心脏保护中的应用提供了可行性。Objective To observe EPO of endotoxin-induced myocardial injury in rats and its receptor expression. Methods Twenty male SD were ran domly divided into LPS group and the control group, LPS group received intraperi- toneal injection of physiological saline, and received intraperitoneal injection of LPS by 10 mg/kg after 3 days of suc- cessive treatment. Then, endotoxin myocardial damage model was established. LPS group and the control group re- ceived equivalent amount of intraperitoneal injection of physiological saline. Bloodletting was carried out 6 hours after intraperitoneal injection from the rat tail and myocardial injury markers were measured. The rat was beheaded alive 6 hours after rat tail bloodletting, its heart tissue was taken out to measure EPO and receptor concentration. Results The serum myocardial injury markers (cTn I, CK, CK-MB) levels for LPS group were significantly higher than those of the control group (t = 6.751, 5.346, 3.432,P 〈 0.01 ). The heart tissue EPO and EPO receptor level for LPS group were also higher than those of the control group (t = 2.587, 4.306, P 〈 0.05). Conclusion There is an existence of EPO and strong EPO receptor expression when there is endotoxin-induced myocardial injury in rats, which proves the feasibility of the application of EPO in sepsis heart protection.
