头孢曲松预处理对局灶性脑缺血大鼠脑组织TLR4和TNF-α蛋白表达的影响

摘　　要：目的探讨头孢曲松钠预处理对局灶性脑缺血大鼠脑组织中Toll样受体4(TLR4)和肿瘤坏死因子(TNF)-α蛋白表达的影响。方法 24只健康SD大鼠随机分为缺血组、假手术组和头孢曲松预处理组。预处理组于术前5 d每日进行腹腔注射头孢曲松钠(200 mg/kg),其余两组给予等量生理盐水腹腔注射。线栓法制备大脑中动脉阻塞(MCAO)模型,缺血后24 h进行神经行为学评分,免疫组化法检测大鼠脑组织TLR4和TNF-α蛋白的表达。结果①神经行为学评分:缺血组神经功能缺损评分较假手术组显著升高(P<0.01);头孢曲松钠预处理组则显著低于缺血组(P<0.05)。②与假手术组比较,缺血组大鼠脑组织的TLR4和TNF-α免疫组化阳性细胞数显著增多(P<0.01);与缺血组比较,头孢曲松预处理组大鼠脑组织的TLR4和TNF-α免疫组化阳性细胞数明显减少(P<0.05)。结论头孢曲松对局灶性脑缺血大鼠脑组织中TLR4和TNF-α的表达有明显的抑制作用,可能是通过抑制脑组织中炎性反应而达到阻断神经损伤的作用。Objective To observe the effect of ceflriaxone sodium on TLR4 and TNF-ct expressions in focal cerebral ischemia rat brain. Methods 24 healthy SD rats were randomly divided into sham-operation, ischemia （model） and ceftriaxone pretreatment groups. In- traluminal filament middle cerebral artery occlusion （MCAO） model of focal cerebral ischemia in rats were established. Ceflriaxone pretreatment group were given intraperitoneal injection of eeftriaxone sodium, 200 mg·kg-1·d^-1 5 days before surgery. 24 h after the focal cere- bral ischemia, the neurological behaviors were evaluated and the expression of TLR4 and TNF-a in hippocampus was determined by immuno- histochemistry. Results ①Neurological deficit scores：compared with that of sham group, the scores of model group evidently increased （P 〈0. 01 ） ; while pretreatment with ceflriaxone significantly decreased neurological deficit scores （P 〈 0. 05 ）. ②Compared with those of sham group, purely cerebral ischemia in rats brain tissue TLR4 and TNF-a positive cell counts were significantly increased （ P 〈 0. 05 ） ; compared with those of model group, eeflriaxone pretreatment brain tissue of rats TLR4 and TNF-a positive cells were significantly reduced （P 〈 0. 05）. Conclusions Ceftriaxone sodium could inhibit the expression of TLR4 and TNF-a in rat brain tissue of focal cerebral ischemia. Ceftriaxone sodium plays an important role in blocking nerve injury through inhibiting inflammatory reaction.
