摘 要:目的:观察海洛因依赖对大鼠颌下腺Bcl-2、Bax阳性细胞表达及分布的影响。方法:正常SD大鼠,随机分为正常对照组和海洛因依赖组,皮下注射海洛因建立海洛因依赖大鼠模型,分别于第10、17、24、31、38天取颌下腺组织,用免疫组织化学Envision法并结合图像分析方法,观察Bcl-和Bax表达。结果:(1)Bcl-2阳性细胞数量在海洛因依赖第10天明显下降(P<0.05),此后开始增加,至第24天时达最高峰(P<0.05),而在第38天时低于正常水平(P<0.05);平均灰度值在依赖第10天明显上升(P<0.05),后开始下降,第24天时达最低值(P<0.05),至第31~38天时接近正常水平(P>0.05);(2)Bax阳性细胞数量在依赖第10天明显降低(P<0.05),第17天时又增加达峰值(P<0.05);之后即开始下降,第31-38天时接近正常水平(P>0.05);其平均灰度值在依赖第10天显著上升(P<0.05),而在第17天时达最低值(P<0.05),后随依赖时间延长,与正常水平无显著性差异(P>0.05)。结论:海洛因对颌下腺Bcl-2、Bax的表达有明显影响,且与依赖时间长短有关。Objective: To observe the effect of heroin dependence on Bcl-2 and Bax positive cells in the submandibular gland of rats. Methods: Normal SD rats were divided into normal control group (NGC) and heroin dependence group (HDG). The heroin dependent model of rats was established by subcutaneous injection of heroin, and submandibular glands were excised on the following 10^th, 17^th, 24^th ,31^st and 38^th days respectively. Immunohistochemical Envision method and image analysis were used in the study. Results: 1. The number of Bcl-2 positive cells descended obviously on the 10th d (P 〈 0.05 ), then, increased and peaked on the 24^th d (P 〈 0.05 ). However, it was lower than that of NCG on the 38^th d (P 〈 0.05 ) ; the mean grey value increased obviously on the 10^th d (P 〈 0.05 ), but decreased and to the lowest on the 24^th d (P 〈0.05). Between the 31^st d and the 38^th d, it ap- proached normal level (P 〉 0.05). 2. The number of Bax positive cells descended obviously on the 10th d (P 〈 0.05 ), then increased and peaked on the 17^th d (P 〈 0.05). Between the 31^st d and the 38^th d, it approached normal level (P 〉 0.05 ) ; The mean grey values increased obviously on thelO^th d (P 〈0.05), but decreased to the lowest on the 17^th d (P 〈0.05). Along with the extension of heroin dependence, it kept the level similar with that of normal (P 〈 0.05 ). Conclusions: Heroin depend- ence affects the expression of Bcl-2 and Bax in submandibular gland, and it relates to the dependence time, which suggests that Bcl-2 and Bax in submandibular gland of rat are involved in regulation process to body injury caused by heroin dependence.
