摘  要:目的:探讨新生大鼠急性肺损伤模型的建立及其病理特征。方法:采用腹腔注射内毒素(LPS,5 mg/kg)的方法制备新生SD大鼠急性肺损伤动物模型,在光镜下观察肺的病理变化,观察支气管肺泡灌洗液(BALF)中细胞分类情况,用ELISA、RT-PCR法检测血清和肺组织中白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的表达。结果:LPS注射3 h后,光镜下可见明显的肺出血和中性粒细胞浸润;肺湿/干重比(W/D)增加,BALF中多形核白细胞(PMN)数量明显增加,血清中TNF-α和IL-6表达水平和肺组织中TNF-αmRNA水平均显著增高。结论:新生SD大鼠腹腔注射LPS后,能够引起显著的急性肺部炎症反应。Objective : To explore the establishment and pathological characteristics of acute lung in- jury model with neonatal rats. Methods: Acute lung injury models of neonatal rats were established via intraperitoneal injection of 5 mg/kg lipopolysaccharides (LPS). The pathological changes were ob- served with tissue sections with microscopy. The tumor necrosis factor-α (TNF-α) mRNA expression in lung were measured by RT-PCR. The levels of TNF-α and IL-6 were detected with ELISA. Re- suits: Lung hemorrhage accompanied by polymorphonuclear neutrophil infiltration was observed in 3 hours after LPS administration. Wet/dry ratio, and polymorphonuclear leukocytes (PMN) in bron- choalveolar lavage fluid (BALF) increased obviously. Serum TNF-α level and IL-6 level significantly increased. TNF-α mRNA expression in lung also increased. Conclusion: LPS administration to neo- natal rats results in acute lung injury accompanied by obvious inflammatory responses.
