摘  要:目的探讨氯诺昔康在大鼠外周神经慢性压迫性损伤模型中对神经行为学改变的影响以及对背根神经节内生长相关蛋白43(growth associated protein 43, GAP-43)和神经生长因子(nerve growth factors,NGF)表达的影响。方法成年SPF级Wistar大鼠50只,按随机数字法分成4大组:正常组(n=5),CCI组,生理盐水对照组(Ns组),氯诺昔康治疗组(L组);CCI组、NS组和L组按术后取材时间分为3、7、14d3个亚组(每亚组5只)。大鼠行右侧坐骨神经慢性压迫性损伤手术,L组的大鼠术后6h腹腔注射氯诺昔康1.3mg/kg,每间隔12h注射一次;CCI组、Ns组和L组的大鼠术后3、7、14d分别测量大鼠的机械缩足反射阈值(mechanical withdrawal threshold, MWT)和热缩腿潜伏期(thermal withdrawal latency,TwL),测量完毕后取出右侧背根神经节,应用免疫组化染色技术研究背根神经节中GAP-43和NGF的表达水平。结果与正常组比较,CCI组、NS组和L组的大鼠术后3、7、14d均出现MWT和TWL明显缩短(P〈0.05),背根神经节内GAP-43和NGF的表达均明显增强(P〈0.05);L组与CCI组比较,术后同时间亚组的MWT和TwL均明显延长(P〈0.05),背根神经节内GAP-43和NGF的表达均明显减少(P〈0.05)。结论腹腔注射环氧化酶抑制剂氯诺昔康能够明显减轻坐骨神经慢性压迫性损伤后的热和机械性痛觉过敏,并使背根神经节内GAP-43和NGF的表达明显减少,说明氯诺昔康对神经性疼痛治疗有效。Objective To investigate the effect of lomoxicam on the neurological behavior change and the expressions of growth associated protein-43 (GAP-43) and nerve growth factor (NGF) in dorsal root ganglion (DRG) on the peripheral nerve chronic constriction injury (CCI) model of rats. Methods Fifty Wistar rats were randomly divided into four groups: normal control group ( n = 5) , CCI model group (CCI group), normal saline control group ( NS group), and lomoxieam therapy group ( L group) ; CCI, NS, and L groups were subdivided into 3 groups according to the different postoperative interval: 3,7 and 14 days ( n =5 each subgroup), respectively. The fight sciatic nerve of rat was to be chronic constriction injure. Group L was given the rat 1.3 mg/kg of lornoxicam every 12 hours; then, the mechanical withdrawal thresh- old (MWT) and thermal withdrawal latency (TWL) of the rats of CCI, NS and L groups were measured at the different postoperative interval (3, 7 and 14 days). After that, the DRG to the injured sciatic nerves were harvested. Immunohistoehemistry was used to examine the expressions of GAP-43 and NGF. Results Comparing with the normal group, the lateneies of MWT and TWL at CCI, NS and L groups were significantly declined at the third, seventh and fourteenth day after surgery ( P 〈 0. 05 ), and the expressions of GAP-43 and NGF in DRG of CCI, NS and L groups were significantly increased after surgery ( P 〈0. 05). Comparing with the CCI group, the MWT and TWL of group L were significantly increased at the same time subgroup ( P 〈0. 05). The expressions of GAP-43 and NGF in DRG of group L were significantly declined at the same time subgroup ( P 〈 0.05 ). Conclusions Lomoxicam could relieve the symptoms of heat and mechanical hyperalgesia after sciatic nerve chronic constriction injury. It proved that lornoxicam was effective in the therapy of neuropathic pain.
