摘 要:乳胶免疫凝集技术是一项成熟的医学检测技术,然而传统检测过程中采用的免疫比浊法都是在宏观条件下进行的,该过程检出限较高且耗液量大。为此,提出一种基于微流控芯片的乳胶免疫凝集光电检测方法。针对乳胶免疫凝集实验过程的特点设计了专用微流控芯片,并创建了具有完整乳胶免疫凝集检测过程(包括进样、混合、反应和检测)的自动化系统平台。以类风湿因子在微流控芯片中的免疫凝集检测为例,对微尺度乳胶免疫凝集系统的最佳条件参数进行了测定与优化。实验结果表明,所设计的基于微流控芯片的乳胶免疫凝集光电检测系统对类风湿因子的检出限比宏观尺度免疫凝集法降低了约60%,且试剂用量是传统免疫比浊法的千分之一。Latex immunoagglutination technique is a mature medical assay technology. However, the immunoturbidimetric method adopted in traditional detection process is carried out under the macroscopic scale condition ,which has the disad- vantages of higher detection limit and reagent consarnption. Therefore, a latex immunoagglutination photoelectric assay method based on microfluidic chip is proposed. Aiming at the features of latex irnmunoagglutination experiment process in microscale,a dedicated microfluidic chip was designed, and an automation system platform with complete latex immunoag- glutination detection process (including sampling,mixing,reacting and detecting) was built. Taking the immunoagglutina- tion detection of rheumatoid factor in microfluidic chip as an example ,the optimum parameters of the latex immunoaggluti- nation system under microscale condition are determined. Experiment results show that the detection limit of rheumatoid factor using the designed latex immunoagglutination photoelectric detection system based on microfluidie chip is decreased by 60% compared with that using the immunoagglutination assay method under macroscopic scale condition. The consumed reagent dosage is one thousandth of that for the conventional immunoturbidimetric assay method.
