摘 要:目的探讨胆管癌细胞株QBC939与正常胆管上皮细胞株HIBEC中微小RNA(miRNA)分子表达谱的差异。方法选取胆管癌细胞株QBC939与正常胆管上皮细胞株HIBEC各6个样本分别作为实验组和对照组,采用miRNA芯片筛选两者之间差异表达的miRNA,样品间表达变化标准以上/下调2倍作为域值范围,并用实时定量聚合酶链反应(Real.timePCR)验证miRNA芯片结果的可靠性。结果在胆管癌细胞株中,上调2倍差异表达miRNA分子72个,上调超过8倍差异表达miRNA6个;下调2倍差异表达miRNA分子61个,下调超过8倍miRNA分子5个(P〈0.05)。结论胆管癌细胞株QBC939与正常胆管上皮细胞株HIBEC之间存在差异表达miRNAs分子,它们可能与胆管癌的发生和转移有关。Objective To explore microRNA differential expression profile between cholangiocar- cinoma cell line QBC939 and normal bile duct cell line HIBEC. Methods Six samples from QBC939 cell line and another 6 samples from normal bile duct cell line HIBEC were chosen as the experimental group and the control group, respectively. MicroRNA profiles in these samples were analyzed by microarray. The threshold value used to screen up- and down-regulated microRNAs was fold chang 〉 2. Real-time quantita- tive polymerase chain reaction ( Real-time PCR) was used to validate the reliability of microarray. Results Six microRNAs were up-regulated in the experimental group and were more than 8 times higher than in the control group. Five microRNAs were down-regulated and were more than 8 times higher than in the control group. Conclusion These microRNAs with differential expression might be related with the formation and metastasis of cholangiocarcinoma.
