性别决定基因相关HMG盒基因4、β-连环蛋白在结直肠癌组织中的表达及意义

摘　　要：目的观察结直肠癌组织中性别决定基因相关HMG盒基因4（Sox4）、β-连环蛋白（β-catenin）的表达，探讨Sox4、β-catenin在结直肠癌组织中表达的意义及其相关性。方法采用免疫组织化学染色方法检测80例结直肠癌组织及20例其邻近正常结直肠组织中Sox4、B—catenin的表达，分析其在结直肠癌组织中的表达与临床病理参数问的关系。结果Sox4在正常结直肠组织黏膜中未见Sox4表达，在结直肠癌组织中高表达，阳性表达率为68．75％（55／80），Sox4在结直肠癌组织中的表达与患者年龄、性别、肿瘤部位、浸润深度、分化程度无明显相关（P〉0．05），Sox4在伴有淋巴结转移、Ⅲ--Ⅳ期的结直肠癌组织中的表达率分别明显高于无淋巴结转移、I～Ⅱ期的表达率（51．2％，P〈0．05）。β-catenin在正常结直肠组织黏膜中主要表达于细胞膜，在结直肠癌组织中异位表达，异位表达阳性率为63．75％（51／80）；B．catenin在结直肠癌组织中异位表达与患者年龄、性别、肿瘤部位、浸润深度无明显相关（P〉0．05）。β-catenin在伴有淋巴结转移、中高分化、Ⅲ一Ⅳ期的结直肠癌组织中的表达率分别明显高于无淋巴结转移、低分化、I一Ⅱ期（P〈0．05）。结直肠癌组织中Sox4表达与β-catenin表达两者呈明显正相关（r=0．501，P〈0．01）。结论Sox4和β-catenin在结直肠癌组织中异常表达，参与结直肠癌的发生、发展，两者之间密切相关。Objective To study the expression and significance of sex determining genes related to HMG box gene 4 （Sox4） and β-catenin in colorectal cancer and their correlation. Methods Immunohisto- chemistry was used to detect the expression of Sox4 and β-eatenin in 80 eases of colorecta] cancer tissues and 20 cases of nearby normal coloreetal tissues, and the relationship between the expression of Sox4 and β-catenin with the clinicopathological parameters was analyzed. Results The Sox4 was mainly expressed in the eell nucleus and the positive rate of Sox4 expresssion in coloreetal cancer was 68.75% （55/80） , but no expression of Sox4 was detected in normal colorectal mucosa. The expression of Sox4 in colorectal cancer was not related to the age, gender, tumor site, the tumor invasion depth and the degree of differentiation （ P 〉 0. 05 ）. The expression of Sox4 was positively associated with lymph node metastasis and elinical stage in colorectal cancer （ P 〈 0. 05 ）. The β-catenin was mainly expressed in the cell membrane in the normal coloreetal mueosa, but it was mainly expressed in the cell plasma in colorectal cancer, and the positive rate of the β-catenin ectopie expression in co]orectal cancer was 63.75% （51/80）. The ectopic expression of B-catenin in colorectal cancer was not related to the age, gender, tumor site, and the tumor invasion depth （ P 〉 0. 05 ）. The expression of β-eatenin was positively associated with lymph node metastasis, histological differentiation, and clinical stage in eolorectal cancer （ P 〈 0. 05 ）. There was a positive correlation between the expression of Sox4 and β-catenin （ r = 0. 501,P 〈 0. 01 ）. Conclusion Sox4 and β-eatenin were abnor- mally expressed in eoloreetal cancer, and they may play important roles in the tumorigenesis and progres- sion of coloreetal cancer and there is a close relationships between them.
