摘 要:目的探讨HSPA2蛋白在食管鳞癌组织中的表达水平及其临床病理意义。方法应用免疫组织化学SABC法检测120例食管鳞癌组织中HSPA2蛋白的表达水平,并分析HSPA2蛋白在食管鳞癌组织中的表达水平与临床病理特征以及患者预后之间的关系。采用实时荧光定量PCR和Western blot检测HSPA2mRNA和蛋白在20例食管鳞癌组织及其配对的癌旁组织和正常组织中的相对表达水平。结果 HSPA2蛋白在120例食管鳞癌组织中的阳性率为75%,其高表达水平与食管鳞癌患者的肿瘤浸润程度、淋巴结转移、TNM分期密切相关(P<0.05);Kaplan-Meier单因素生存分析显示HSPA2蛋白高表达组患者的5年无病生存率(7.8%)和总生存率(18.9%)显著小于低表达组患者的40%和66.7%(P<0.05);Cox回归模型显示HSPA2蛋白表达在无病生存率和总生存率中危险比为2.577和2.899,是食管鳞癌患者的一个独立危险因素(P<0.05)。与正常食管上皮组织和癌旁组织相比,HSPA2在食管鳞癌组织中mRNA表达水平较正常食管上皮组织显著上调8.71倍,较癌旁组织上调2.12倍。HSPA2蛋白在食管鳞癌组织(0.44±0.15)中表达较配对的癌旁组织(0.20±0.06)和正常组织(0.11±0.06)中表达亦明显增高(P<0.05)。结论 HSPA2在人类食管鳞癌中高表达,其在食管鳞癌的发生和发展过程中起重要作用,与肿瘤侵袭及转移密切相关。【Objective】 To explore the expression and clinical significance of HSPA2 in esophageal squamous cancer.【Methods】 The expression of HSPA2 protein in 120 primary esophageal squamous cancer tissues specimens was analyzed by immunohistochemical SABC staining,meanwhile which was correlated with clinicopathological parameters and prognosis of patients.Additionally,the expression of HSPA2 mRNA and protein in 20 primary esophageal squamous cancer tissues,paired 20 adjacent non-cancer tissues and 20 normal tissues were evaluated by real-time quantitative PCR and Western blot respectively.【Results】 The positive rate of HSPA2 protein was 75%,and the high expression of HSPA2 protein was significantly associated with depth of invasion(T),lymph node metastasis(N),clinical TNM stage respectively(P &lt;0.05).Kaplan-Meier univariate survival analysis showed that five-year Disease-free Survival and Overall Survival rates for patients with high expression of HSPA2 protein were 7.8% and 18.9%,which were significantly lower than 40% and 66.7% in patients with low expression of HSPA2 protein(P &lt;0.05).Multivariate Cox regression analysis revealed that HSPA2 expression was an independent prognostic factor in Disease-free Survival and Overall Survival for patients with esophageal squamous cancer(Hazard ratio was 2.577 and 2.899 respectively,P &lt;0.05).Compared with normal tissues and adjacent non-cancer tissues,the expression of HSPA2 mRNA in 20 primary esophageal squamous cancer tissues were significantly increased by 8.71 times and 2.12 times separately.The expression of HSPA2 protein in esophageal squamous cancer tissues(0.44±0.15) was also significantly higher than that in paired adjacent non-cancer tissues(0.20±0.06) and normal tissues(0.11±0.06)(P &lt;0.05).【Conclusion】 Our findings suggested that HSPA2 was high expression in esophageal squamous cancer,which might have played an important role in the malignant tumorigenesis and progression of esophageal squamous can
